The Rab7 C-terminal intrinsically disordered region is a key determinant of development and pathogenicity in Aspergillus flavus.

The findings extend the mechanistic understanding of Rab7 beyond its catalytic pocket and establish the AflRab7 C-terminal IDR as a critical regulatory element that couples vacuolar dynamics to A. flavus development and aflatoxin biosynthesis, highlighting a tractable target for mitigating fungal toxigenic potential.

Enterovirus 2B protein: From viroporin to membrane stress sensor, mechanistic principles of multi-pathway regulation

ABSTRACT Enterovirus 2B is a highly conserved, functionally versatile, nonstructural protein. The classic viroporin model, which restricts its role to ion channel formation, cannot account for its interactions with host factors such as Bcl-2-associated X protein, interleukin enhancer-binding factor 2, and karyopherin s...

Anagrelide remodels the PDE3A-SLFN12 interactome to associate with translation initiation machinery in cancer cell lines.

In this proposed model, anagrelide remodels the interactome of the PDE3A-SLFN12 complex, which relocalizes and accumulates near the mRNA entry channel of the 43S pre-initiation complex, which provides important mechanistic context for how PDE3A modulators act within the cell.

Identification of TMEM263 as an ER-resident protein that controls early stages of lipid droplet biogenesis

It is proposed that TMEM263 can interact with and support the condensation of neutral lipids in a bilayer to promote lipid droplet formation and growth and provide a mechanistic understanding linking these phenotypes to impaired lipid droplet biology.

TgDDP is a novel DIX domain protein that plays an essential role in Toxoplasma endodyogeny

ABSTRACT Apicomplexan parasites are protozoan pathogens responsible for major human diseases, including toxoplasmosis, malaria, and cryptosporidiosis. Toxoplasma gondii has emerged as a model for studying cell division in tissue coccidians. Unlike higher eukaryotes that divide by binary fission, Toxoplasma replicates v...

Cell surface remodeling caused by the loss of the flippase subunit TMEM30A in immune cells

Using a live-cell lipid reporter, membrane order probe, and surface proteome mapping, it is shown that TMEM30A-knockout cells display robust PS externalization accompanied by faster lateral diffusion of membrane constituents and decreased plasma membrane order.
