Abstract
The use of herbicides represents one of the effective weed management strategies. To create high-efficiency, low-risk protoporphyrinogen IX oxidase (PPO)-targeted herbicides, a set of novel N-phenylphthalimides bearing 1,3,4-oxadiazole and acylhydrazide fragments was designed and synthesized. Bioassays revealed that most of the compounds possessed potent herbicidal activity against six common weeds. Compound A9 exerted good inhibitory effects on Nicotiana tabacum PPO (NtPPO) with a Ki of 14.0 nM, outperforming commercial flumioxazin (46.3 nM) and flumiclorac-pentyl (52.0 nM). Molecular docking validated that A9 binds tightly to NtPPO via hydrogen bonds with Arg98 (2.8 Å), π-π stacking with Phe392, and π-alkyl sandwich interactions with Leu356/Leu372. Assays with PPO-deficient and PPO-complemented Escherichia coli strains, alongside AtPPO-overexpressing and mutant Arabidopsis, collectively verified A9 functions as a target-specific PPO inhibitor.
Protoporphyrinogen oxidase (PPO) is one of the major targets for herbicide discovery. N-Phenylpyrazole derivatives bearing ester, oxime, and isoxazoline moieties were synthesized via isomerization, derivatization and cyclization. Bioassays showed C34 potently inhibited Nicotiana tabacum PPO (NtPPO) (Ki = 15.6 nM), wi...

Protoporphyrinogen IX oxidase (PPO) is a validated target for fast-acting herbicides. To identify low-dose PPO-inhibiting leads, 41 benzyl-substituted pyrimidinedione derivatives were designed by side-chain optimization of epyrifenacil and synthesized. In greenhouse postemergence screening, FM-604 provided 100% contr...

To continue our research on developing PPO inhibitors, we replaced the triazinone core of compounds B, G, and H from our previous work with pyrimidinediones, employing a scaffold-hopping strategy to design and synthesize compounds I, J, and K. Compound I18 exhibited potent NtPPO inhibition (Ki = 10.9 nM) and broad-sp...

4-Hydroxyphenylpyruvate dioxygenase (HPPD) is an attractive target for the development of bleaching herbicides. To develop novel HPPD inhibitors, 40 diketonitrile derivatives were designed by active-fragment splicing, synthesized, and characterized. In vitro inhibitory assays against Echinochloa crus-galli HPPD (EcHPPD...

4-Hydroxyphenylpyruvate dioxygenase (HPPD) has become an attractive target for herbicide development owing to the slow development and low risk of resistance to its inhibitors. However, most available HPPD inhibitors exhibit limited crop selectivity and poor efficacy against grasses. Therefore, the exploration of novel...

Transketolase (TKL; EC 2.2.1.1) has been validated as a potential new herbicide target. To develop a new and highly effective herbicides-targeted TKL, a series of indole-containing fusarium acid (FA) derivatives 9a-9s were designed and synthesized. Among them, compound 9o exhibited more excellent herbicidal activity ag...
