Because BaP may not fully account for the toxicity of all PAHs, this study provides relevant quantitative benchmarks for intestinal toxicity, advancing knowledge on current PAH exposure risk assessment.

Abstract

Despite their well-established carcinogenicity, the effects of dietary polycyclic aromatic hydrocarbons (PAHs) on intestinal cells remain poorly understood. Additionally, toxicity studies are often based on animal models, and interspecies differences in xenobiotic metabolism may limit extrapolation to human exposure. The cytotoxic and toxicogenomic effects of seven food-relevant PAHs-benz[a]anthracene (BAA), benzo[a]pyrene (BaP), benzo[b]fluoranthene (BBF), benzo[k]fluoranthene (BKF), chrysene (CHRY), indeno[1,2,3-cd]pyrene (INDE), and dibenz[a,h]anthracene (DBAHA)-were investigated in mouse (C57BL/6), pig (IPEC-J2), and human (HIEC-6) intestinal epithelial cells. Viability assays showed that the LC50 values of the PAHs for the mouse and pig models were 5-7 and 6-9 μg/mL, respectively, compared with 12-17 μg/mL in human cells. A conserved transcriptional response to PAH exposure was observed across all cell lines, including upregulation of xenobiotic metabolism genes (ahr, cyp1a1, cyp1b1, gst) and proliferation gene marker (ki67), and downregulation of barrier and junction genes (muc2, cdh1). PAH-treated HIEC-6 cells showed metabolic activation, with cyp1a1 and cyp1b1 upregulated more than 32-fold. Immunofluorescence microscopy experiments supported our findings of decreased MUC2 protein after PAH treatment. Because BaP may not fully account for the toxicity of all PAHs, our study provides relevant quantitative benchmarks for intestinal toxicity, advancing knowledge on current PAH exposure risk assessment.

Genotoxic and cytokine-associated responses to benzo[g,h,i]perylene and benzo[b]fluoranthene in a human lung co-culture model at the pseudo-air-liquid interface.

Polycyclic aromatic hydrocarbons (PAHs), including benzo[g,h,i]perylene (BGP) and benzo[b]fluoranthene (BBF), are ubiquitous airborne pollutants with significant toxicological relevance. Inhalation represents the primary exposure route, with the respiratory epithelium and alveolar macrophages serving as key targets for...

Cytotoxic and Oxidative Effects of Common Pesticides on Hepatic and Renal Cell Lines (HepG2 and Hek-293): Implications for Human Health and Environmental Safety.

Modern agricultural practices expose humans and the environment to numerous chemical pesticides. Increasing evidence suggests that commercial pesticide formulations may exhibit toxicological properties that differ from those of their active ingredients alone due to the presence of co-formulants and additives. This stud...

Chronic co-exposure to benzo[a]pyrene and BDE-47 induces malignant transformation of human mammary epithelial cells via dysregulation of the miR-491-5p/VPS28 axis.

Benzo[a]pyrene (B[a]P) and 2,2',4,4'-tetrabromodiphenyl ether (BDE-47) are widespread environmental pollutants associated with breast carcinogenesis, yet their combined effects remain unclear. In this study, a chronic low-dose exposure model was established using human mammary epithelial MCF-10A cells to investigate co...

The Influence of a Human-Relevant Polychlorinated Biphenyl Mixture on the Liver in Male and Female Mice

Polychlorinated biphenyls (PCBs) are persistent environmental contaminants associated with hepatic toxicity, yet the biological effects of exposure to complex, environmentally relevant PCB mixtures remain incompletely defined. This study evaluated hepatic accumulation and molecular responses to oral exposure to the H...

Benzo[a]pyrene-induced immunotoxicity, neurotoxicity, and cardiotoxicity in grass carp (Ctenopharyngodon idella)

Introduction Benzo[a]pyrene (B[a]P), a widespread polycyclic aromatic hydrocarbon (PAH), is a major aquatic pollutant with known toxic effects. However, its immunotoxicity during early fish development remains poorly understood. This study investigated the immunotoxic, neurotoxic, and cardiotoxic effects of B[a]P in gr...

Benzo[a]pyrene and bisphenol A interact to induce combined pulmonary toxicity through an integrated transcriptomic-metabolomic network.

Widespread atmospheric pollutants such as benzo[a]pyrene (BaP) and bisphenol A (BPA) persistently coexist, yet their combined risks to the respiratory system remain largely overlooked. In this study, C57BL/6J mouse and A549 cell models were employed to investigate the combined pulmonary toxicity induced by BaP and BPA,...
