TL;DR

Using Seqtometry-based analysis of mouse and human cDCs, distinct steady-state cDC1 gene programs enriched for either tolerance-associated or immune response-associated (pre-immunogenic) transcriptomic features that are present under homeostatic conditions in fully differentiated cDCs with divergent predicted immune functions are identified.

Inflamed Yet Immune-Evasive? A Transcriptomic Meta-Analysis Identifies Conserved Inflammatory, Developmental, and Neuronal Signatures Associated with Polyploid Giant Cancer Cells

This study proposes that PGCCs represent an immune-adaptive polyploid survival state in which inflammatory and ontogenetic pathways are repurposed to support immune evasion and tumor persistence and provides a framework for therapeutic strategies aimed at dismantling the PGCC reservoir and preventing tumor relapse.

An integrated cell atlas of the innate lymphoid cells in health and disease 2255047

Innate lymphoid cells (ILCs) represent a highly diverse family of lymphocytes that play pivotal roles in tissue homeostasis, inflammation, and cancer immunity. Despite growing evidence of their plasticity and functional heterogeneity, a comprehensive understanding of human ILC diversity across pathological contex...

A human cytokine response atlas to reconstruct underlying gene regulatory networks

Circulating cytokines encode immune state, yet their pleiotropy and cell-type specificity make constructing a unified atlas of immune cell responses to them challenging. Here, I transformed a single-cell atlas of approximately 10 million human peripheral blood mononuclear cells from 12 donors exposed to 90 cytokines in...

Inflammatory hematopoiesis drives intestinal inflammation and is modulated by anti-TNF therapy 2309660

Chronic inflammatory diseases often originate in local tissues yet display systemic immune abnormalities, suggesting durable changes in upstream immune regulation. Crohn’s disease (CD), while centered in the intestine, is a systemic inflammatory disorder. Hematopoietic stem and progenitor cells (HSPCs) can retain...

Comparative Transcriptomic Profiling in Two Widely Used Human Cell Lines Delineates Their Shared and Distinct Patterns of Interferon Responses

Simple Summary Interferons (IFNs) are critical for pathogen resistance and immune modulation. While IFN response occurs in many cell types, a high-resolution comparison across cell types is lacking. By using RNA sequencing, this study conducted a time-serial charting of the transcriptomic responses in two widely used i...
