P1.109. Overexpression of RFC3 Relates to Tumor Development and Poor Outcome of Esophageal Squamous Cell Carcinoma

Abstract

Esophageal Cancer: Molecular Biology/Pathology Replication factor C (activator 1) 3 (RFC3), located on 13q13.2, is reported to be necessary for the elongation of primed DNA templates by DNA polymerase delta and DNA polymerase epsilon. In this study, we tested whether RFC3 acts as a cancer-promoting factor by being overexpressed in esophageal squamous cell carcinoma (ESCC). We analyzed 14 ESCC cell lines and 58 primary tumor samples of ESCC, which were curatively resected in our hospital. RFC3 protein was detected in all cell lines and primary tumor samples (39/58 cases; 67.2%). Knockdown of RFC3 using specific siRNA inhibited proliferation in TE-5 and TE-9 of which RFC3 overexpressed strongly. Overexpression of RFC3 protein correlated with lymphatic invasion and worth prognosis. Moreover, multivariate analysis demonstrated that RFC3 was independently associated with worse outcome (p= 0.027, hazard ratio 3.4 (11-15)). These findings suggest that RFC3 plays a crucial role in tumor cell proliferation by its overexpression and highlight its usefulness as a prognostic factor and potential therapeutic target in ESCC.
