Abstract

This study evaluated bromelain-induced structural changes in beef myofibrillar proteins (MP) and explored related molecular mechanisms. Bromelain treatment markedly changed MP physicochemical features and structural conformation, facilitated aggregate dissociation, and increased dispersion and solubility (P < 0.05). The 0.50% bromelain treatment showed the greatest effect, increasing MP solubility to 81.53% and reducing the PDI to 0.338 ± 0.036. It also exposed previously buried hydrophobic moieties and thiol groups, reduced the proportion of ordered structures, and shifted MP from a compact, ordered conformation toward a loose, disordered state. Molecular docking indicated that bromelain bound stably to myosin heavy chain (MHC), primarily via hydrogen-bond interactions, with a binding energy of −12.4 kcal/mol. Molecular dynamics simulations further revealed that bromelain binding reduced the structural stability of MHC and promoted a looser conformation. These results offered theoretical support for employing bromelain to improve meat tenderness and regulate product quality.

Effects of Maillard glycosylation with different saccharides on structural, antioxidant and functional properties of porcine myosin.

Myosin is a crucial protein that contributes to the processing performance of meat products. This study investigated the effects of Maillard glycosylation with glucose, sucrose, and β-glucan on the structural, antioxidant, and functional properties of porcine myosin. During 0-8 h of reaction, the glycosylation degree,...

Staged structural changes of fermented chicken breast myofibrillar protein: Conformational unfolding and aggregation.

Fermented chicken breast is promoted as a healthier alternative to traditional high-fat fermented meat products, yet its quality depends critically on the structural state of myofibrillar protein (MP). To clarify how MP deteriorates during extended natural fermentation, this study established a 150-day fermentation mod...

High Hydrostatic Pressure: A Tool for Functional Modification of Lupin Protein Isolates

This study aimed to evaluate high hydrostatic pressure (HHP) processing as a modification method for protein functionalization. Lupin protein isolates (LPI), derived from the species Lupinus luteus, were subjected to pressures of 200, 350, and 500 MPa for 10 min at 20 °C, and the impact of HHP on protein structure and...

Changes in the Molecular Forces and Rheological Properties of Pork Myosin with Sodium Bicarbonate

To investigate how sodium bicarbonate concentrations (0, 10, 20, 30, 40, and 50 mM) modulate the turbidity and molecular interactions of pork myosin, the pH, turbidity, particle size, active and total thiol content, surface hydrophobicity, rheology properties, and chemical forces were analyzed. Results revealed that in...

Interactions between Myofibrillar Proteins and Quercetin: Underlying Mechanisms and Effects on Interfacial Film Formation and Emulsion Stability.

Mechanisms underlying myofibrillar protein (MP)-quercetin interactions and the effects of these interactions on interfacial film formation and MP emulsification properties were investigated. These interactions represent an enthalpy-driven, spontaneous, and exothermic process predominantly driven by hydrogen bonds, van...

Multispectroscopic and molecular simulation insights into the interaction between casein and key aroma compounds in yogurt.

The molecular mechanisms governing interactions between casein (CS) and key yogurt aromas remain insufficiently elucidated. This study systematically investigated the interactions of CS with acetoin, diacetyl, and acetaldehyde through multispectroscopic techniques and integrated molecular simulations. Fluorescence quen...
