Catalytic asymmetric dearomative cycloaddition provides a powerful route to polycyclic chiral frameworks, yet the precise control of selectivity in such cycloadditions remains a formidable challenge. Herein we report a mechanism-guided strategy that harnesses the unique reactivity of vinylidene

-quinone methides as highly reactive and multifunctional synthons to realize such cycloaddition with diverse (hetero)polycyclic aromatics. Density functional theory calculations revealed that the distinctive charge distribution, orbital delocalization, and narrow frontier molecular orbital gap of the synthons underpin their multisite reactivity and predictable selectivity. Leveraging these features, we accomplished cycloadditions at challenging sites with excellent regio-, chemo-, and enantioselectivity, including the C6a–C13c site of dibenzo[

The direct addition of C–H bonds across allenes represents an attractive strategy for the construction of allylic frameworks, but controlling the selectivity remains a major challenge. In this Synpacts article, we summarize the development of an Ir(I) catalytic system featuring a perfluorinated bisphosphine ligand, d F...

Chiral spirocycles exhibit tremendous potential in drug discovery and asymmetric synthesis, owing to their unique three-dimensional rigid framework and superior stability. However, stereoselective construction of polycyclic spiro compounds through a single catalytic transformation remains a significant and enduring cha...

Achieving general asymmetric catalytic manifolds that accommodate broad substrate diversity remains a long-standing and highly sought-after goal in synthetic chemistry. Herein, we report an efficient cobalt-catalyzed desymmetric hydrogenation of structurally diverse cyclic 1,2- and 1,3-diacid anhydrides, enabling acces...

Thiazoles are ubiquitous motifs in bioactive molecules, making strategies for the stereospecific installation of chiral C(sp3)-rich substituents on this scaffold particularly valuable for drug discovery. However, achieving such transformations remains nontrivial, as conventional C(sp2)-C(sp3) cross-coupling approaches...

Chiral medium-sized rings, defined herein as 7- to 11-membered cyclic frameworks, are common in bioactive molecules and occupy an important but still underexplored region of drug-like chemical space. Their asymmetric synthesis remains formidably challenging, as conformational flexibility, transannular interactions, and...
