Chiral spirocycles exhibit tremendous potential in drug discovery and asymmetric synthesis, owing to their unique three-dimensional rigid framework and superior stability. However, stereoselective construction of polycyclic spiro compounds through a single catalytic transformation remains a significant and enduring challenge, owing to steric hindrance and electronic effects. Here we report a Pd/Sadphos-catalyzed dynamic kinetic asymmetric cascade dearomative Heck/Tsuji–Trost reaction of racemic phenolic biaryls with 1,3-dienes, enabled by transient axial-to-point chirality induction and transfer via a dynamic kinetic asymmetric transformation (DyKAT) manifold. Over 40 valuable polycyclic spiro enones bearing three contiguous tertiary/quaternary stereocenters are afforded in moderate to high yields, with excellent regio-, diastereo- and enantioselectivity (up to 99% ee). This work provides a modular synthetic strategy for complex chiral spirocycles and expands the scope of transient chirality transfer chemistry. Chiral polycyclic spirocycles are privileged molecular scaffolds for drug discovery and asymmetric synthesis, yet their efficient stereoselective construction remains challenging. Here, the authors report a Pd/Sadphos-catalyzed asymmetric dearomative cascade that delivers polycyclic spiro enones with three contiguous stereocenters and exceptional stereoselectivity.
Inherently chiral molecules generated by the curvature of planar frameworks represent an emerging class of three-dimensional chiral architectures, yet their catalytic asymmetric synthesis remains highly challenging. Herein, we report a Pd-catalyzed asymmetric Suzuki-Miyaura coupling for the efficient construction of in...
Dynamic kinetic resolution (DKR) serves as a fundamental approach in asymmetric synthesis which enables the efficient conversion of racemic starting materials into a single stereoisomer, yet its extension to racemic diastereomers remains a formidable challenge, and reported DKR processes have largely been restricted to...
We report a regiodivergent strategy for the catalytic asymmetric synthesis of enantioenriched spirocyclic γ- and δ-lactams from a single cyclic alkyne-tethered β-ketoamide precursor. Critically, simply switching the catalyst system between Cu(I)/Pyrabox and Ni(II)/Box enables precise toggling of the cyclization between...

There has been a recent surge in research aimed at synthesizing inherently chiral macrocycles that lack point, axial, planar, or helical chirality. To overcome the structural limitations of existing inherently chiral macrocyclic systems, which often suffer from facile racemization due to conformational flexibility, we...

Catalytic asymmetric dearomative cycloaddition provides a powerful route to polycyclic chiral frameworks, yet the precise control of selectivity in such cycloadditions remains a formidable challenge. Herein we report a mechanism-guided strategy that harnesses the unique reactivity of vinylidene
