TL;DR

This study identifies the NSP1β/NSP2 and ORF1b/ORF2 sites as suitable and genetically stable insertion points for foreign genes within the NADC30-like PRRSV genome, providing an experimental basis for optimizing PRRSV-based foreign gene expression vectors and developing multi-gene expression platforms.

Abstract

Porcine reproductive and respiratory syndrome virus (PRRSV) is a major pathogen threatening the global swine industry. Infectious clone and recombinant virus platforms serve as vital tools for investigating PRRSV biology and developing genetically engineered vaccines. In this study, we constructed a full-length infectious clone of the NADC30-like PRRSV strain GX-2019 and systematically evaluated the tolerance of various genomic regions to foreign gene insertion. We introduced an EGFP expression cassette into selected genomic junction sites and assessed the rescue efficiency, foreign gene stability, and replication characteristics of the recombinant viruses. Among the structural protein regions, recombinant viruses with EGFP insertions at the ORF1b/ORF2 and ORF4/ORF5a junctions were successfully rescued; among the 12 junction regions within the non-structural protein coding sequences, the NSP1β/NSP2 junction was the only site that supported stable foreign gene expression. The replication kinetics and plaque phenotypes of these viruses were comparable to those of the parental virus. Serial passage analysis demonstrated that both the NSP1β/NSP2 and ORF1b/ORF2 sites maintained good genetic stability for the inserted foreign genes. Furthermore, by utilizing the NSP1β/NSP2 and ORF1b/ORF2 sites as independent insertion points for EGFP and mCherry expression cassettes, respectively, we constructed a dual-fluorescence recombinant PRRSV capable of simultaneously and stably expressing both EGFP and mCherry.

A newly established reverse genetic system for a circular RNA virus reveals new requirements for infection and its biocontrol potential

This work constructed the first infectious cDNA clone of an ambivirus, Fusarium graminearum ambivirus 1 (FgAV1), using a head-to-tail dimer placed downstream of a fungal promoter, demonstrating that FgAV1 infection triggers a fungal RNAi response, extending the antiviral role of host sRNAs to circular RNA viruses.

Genetic characterization of Orf virus isolates targeting PACR gene: Optimization of a diagnostic PCR for host-specific differentiation of goat and sheep origin isolates.

Parapoxviruses are known for its unique feature of carrying a number of virulence genes in their large genome and deploy them as a weapon to evade host immune response. One of its unique gene is Poxviral anaphase promoting complex regulator (PACR) from the left terminal of the genome named as ORF014. This PACR protein...

Generation and characterization of a PRRSV-2 DIVA marker virus by deletion of a novel epitope in the nucleocapsid protein

The feasibility of developing a PRRSV DIVA vaccine through targeted modification of an N protein epitope is demonstrated, providing a potential strategy to facilitate PRRS control and suggests that combining the recombinant virus with cELISA diagnostic tests allows accurate differentiation between vaccinated and wild-t...

Reverse genetics system for emerging tick-borne orthonairovirus highlights dispensable N-terminal region of Gn associated with replication in tick vectors

A growing number of emerging and re-emerging tick-borne orthonairoviruses belonging to the Sulina and Tamdy genogroups have recently been identified in association with febrile diseases in East Asia. Yezo virus (YEZV) is one such viruses and is genetically distinct from well- characterized Crimean-Congo hemorrhagic fev...

Construction of Recombinant PRV Expressing PRRSV Antigens and Evaluation of Their Protective Efficacy in Pigs Against Homologous Challenge

Background/Objectives: Porcine reproductive and respiratory syndrome virus (PRRSV) remains a major threat to the global swine industry, while the emergence of variant pseudorabies virus (PRV) strains has further complicated disease control in China. This study aimed to develop a PRV-based viral vector platform for PRRS...
