Immunogen selection and prior immunity shape antibody breadth following immunisation with avian H5 hemagglutinin

Avian influenza A viruses pose a persistent zoonotic threat to humans owing to their expanding host range and high case fatality rates. In particular, viruses from the 2.3.4.4b clade of the H5 subtype have now been detected in over 60 mammalian species, raising serious pandemic concerns. Understanding immune recognitio...

A dimeric mRNA vaccine based on key-mutant and KP.3 RBDs induces broad-spectrum immunity and protects mice against JN.1 and XDV challenges

The continued antigenic evolution of SARS-CoV-2 Omicron subvariants has progressively eroded vaccine-elicited protective immunity, driving demand for next-generation candidates that confer broad-spectrum protection against phylogenetically divergent strains. Here we report the design and preclinical evaluation of SV, a...

Convergent IGHV3-53/3-66 antibodies elicited by Omicron BA.1 infection broadly neutralize emerging SARS-CoV-2 variants

G germline revertant analysis of the COV2-3731 mAb provided additional insights into how this COV2-3731 and other IGHV3-53/3-66-encoded public antibodies evolve to gain breadth against antigenically distinct SARS-CoV-2 variants such as BA.2.3.2, BA.3.1, KP.2.2, and, to some extent, KP.3.3.2.

Cross-Neutralizing Antibody Responses to Diverse Coronaviruses in the Human Population.

It is demonstrated that post-pandemic sera exhibit enhanced cross-neutralizing antibody responses predominantly against sarbecoviruses, potentially strengthening population immunity against related zoonotic sarbecoviruses and increasing the immunological barrier to their emergence in humans.

Durability and Breadth of Neutralizing Antibodies Against SARS-CoV-2 Variants Following XBB.1.5 Vaccination in a Multiply Exposed Cohort.

The observed waning of cross-neutralizing antibodies against emerging variants underscores the challenge of maintaining durable protection and supports the need for continued monitoring of antibody responses to guide evidence-based updates to COVID-19 vaccines.

Complement augments antibody neutralization of SARS-CoV-2 variants.

The data show that complement potentiates the ability of antispike antibodies to neutralize immune-evasive SARS-CoV-2 variants and in sera from individuals with severe COVID-19, both complement-mediated neutralization and anti-S2 antibody levels were associated with improved survival.
