STAT1 gain-of-function variant in a previously healthy patient with disseminated Mycobacterium genavense infection

STAT1 defects present with diverse clinical phenotypes. Thiagarajan et al. describe an unusual genotype/phenotype: disseminated mycobacterial infection without CMC in the context of a pathogenic STAT1 GOF variant (p.Lys388Glu).

Lifelong Genetic Inhibition of PCSK9 and Hepatic Safety

This genetic association study investigates whether the full spectrum of rare PCSK9 loss-of-function variants is associated with incident liver-related outcomes, given the development of therapies targeting proprotein convertase subtilisin/kexin type 9 (PCSK9).

Monoallelic SH2B1 Variants in Neurodevelopmental Disorders: Phenotypic Expansion and Functional Characterization of Rare Variants

SH2B1 encodes an adaptor protein involved in metabolic regulation and body‐weight control. Although SH2B1 haploinsufficiency is a recognized contributor to obesity in 16p11.2 deletion syndrome, the relationship between rare monoallelic SH2B1 variants and neurodevelopmental disorders (NDDs) remains poorly characterized.

Celiac disease and developmental delay as the first manifestations of tripeptidyl peptidase 2 deficiency

shift toward a senescent B-cell profile may represent an important feature of the immune dysregulation observed in our patient. The combination of reduced functional B-cell subsets and underlying immune senescence likely explains the patient's susceptibility to recurrent otitis media and pneumonia despite complete diet...

Mosaic TLR8 gain-of-function in an elderly female treated with JAK inhibitor and adjunctive therapy

Patel et al. describe an elderly woman with severe chronic neutropenia caused by mosaic TLR8 gain-of-function. The report broadens a primarily pediatric, male disease spectrum and highlights somatic mosaicism as an important diagnostic consideration in adults with unexplained cytopenia and immune dysregulation.
