These findings define a paradigm for intracellular channel regulation and offer a structural blueprint for designing specific inhibitors of TASK-2.
Abstract
Significance The potassium channel TWIK-related acid-sensitive K+ channel 2 (TASK-2) is a critical physiological sensor that regulates breathing and kidney function to maintain pH homeostasis. Despite its importance, the molecular mechanisms controlling its intracellular gating and inhibition have remained unclear. We use cryoelectron microscopy and electrophysiology to address these questions. We found TASK-2 employs a unique mechanism to gate closed in response to low intracellular pH that involves protein and lipid components. We further show that the local anesthetic bupivacaine inhibits TASK-2 by binding inside the channel at a site that overlaps with the lipid binding site. These findings define a paradigm for intracellular channel regulation and offer a structural blueprint for designing specific inhibitors of TASK-2.
BK channels are a class of K+ channels that possess an unusually high conductance and are synergistically gated by intracellular Ca2+ and voltage. Despite the significant array of experimental and computational data, many aspects of their function and dynamics remain unclear - such as how ion permeation is halted in th...

Inhibition of Kv channels by 4-aminopyridine (4AP) improves motor function in multiple sclerosis by enhancing neuronal excitability. The mechanism of inhibition and the structural basis of 4AP binding to Kv channels remain unclear. Here, we determined the structure of the Shaker V369I-I372L-S376T (ILT) mutant bound to...

It is argued that the central challenge is no longer simply to activate or inhibit TRPM4, but to stabilize the right conformational state in the right tissue at the right stage of pathology.

Significance Developing drugs that selectively target closely related ion channel isoforms remains a major challenge because these proteins often share highly conserved drug-binding sites. Here, we show that selective activation of Kv7 potassium channel isoforms by the rosemary-derived metabolite carnosic acid (CA) ari...

This work identifies a conserved gating crosstalk mechanism linking the activation gate to the width of the selectivity filter entrance, which regulates ion permeation rate by tuning ion occupancy at the selectivity filter.
