Abstract

Simple Summary This study presents an integrative proteomic analysis of endothelial cells, pericytes, and astrocytes—the three main cell types orchestrating vascular and cerebral metabolism and functions—using whole-cell lysates and subcellular fractionation combined with data-dependent acquisition mass spectrometry. Subcellular fractionation enhanced proteome coverage by up to 93%, especially for membrane-associated and insoluble proteins, revealing sharper cell-type-specific functional signatures. However, sampling strategy and cell differentiation status were shown to influence protein detection and interpretation of specificity. By merging data across approaches, we generated reference proteomes for each cell type, offering valuable resources for neurovascular research and biomarker discovery.

VINE-seq and MultiVINE-seq for single-nucleus and multiome profiling of the brain vasculature.

VINE-seq provides a robust, reproducible workflow for the enrichment and high-resolution profiling of vascular, perivascular, and immune cells from fresh or frozen human and mouse brain tissue and optimized extraction of nuclei from purified vessels using enzymatic digestion.

Proteomics analysis of small extracellular vesicles derived from human retinal Müller glial cells under high glucose condition.

Diabetic retinopathy (DR) is a leading cause of vision loss and is characterized by early dysfunction of the retinal neurovascular unit (NVU), preceding neurodegenerative and vascular pathology. Müller glial cells are central regulators of retinal homeostasis, but the contribution of Müller glia-derived small extracell...

Mitochondrial Molecular Diversity in the Brain

These findings challenge the conventional view of mitochondria as generic metabolic engines and position mitochondrial molecular diversity as a fundamental feature of brain organization, with direct relevance to behavior, aging, and neurological disease.

Protocol for integrated phosphoproteomic and proteomic analysis of the mouse ventral tegmental area with functional and kinase motif enrichment.

We present a protocol for label-free proteomic and phosphoproteomic analysis of the mouse ventral tegmental area (VTA). We describe steps for combining region-specific biopsy-punch collection, parallel proteomic and phosphoproteomic processing, liquid chromatography-tandem mass spectrometry (LC-MS/MS), and downstream e...
