Abstract

The formidable biofilm-forming ability of Pseudomonas fluorescens enables its persistent contamination under distribution conditions (0 - 4℃) of chilled meat, yet the mechanisms governing its biofilm formation remain inadequately explored. In this study, with 25℃ serving as the control, P. fluorescens developed more substantial and architecturally coherent biofilms at 4℃ during 96 h-incubation, accompanied by increased extracellular polymeric substances synthesis. This response phenotype enhanced cell adhesion to facilitate persistence and increased the cross-contamination after multiple cell transfers. Transcriptomic analysis revealed that 514 differentially expressed genes (DEGs) were up-regulated and 327 DEGs were down-regulated, which were involved in fatty acid degradation, flagellar assembly, lysine degradation, pyruvate metabolism, etc. Notably, as primary regulatory pathways, flagellar assembly and fatty acid metabolism suggested they could facilitate microbial accumulation and the efficient conversion of fatty acids, respectively. Correspondingly, changes in flagellar assembly-related genes and fatty acid degradation-related genes were consistent with transcriptome results. In view of these, the apparent reduction in motility and the increase in unsaturated fatty acid content, along with the enhancement of cell surface hydrophobicity and auto-aggregation, further verified the above-mentioned pathways. This study provided insights into formation mechanism of P. fluorescens biofilm and the development of control strategies.

Uncovering the phenotypic and transcriptional characteristics of biofilm-released cells in Pseudomonas aeruginosa

Pseudomonas aeruginosa is a prominent biofilm-forming pathogen associated with chronic infections that are difficult to eradicate due to the protective biofilm matrix. Inducing biofilm dispersion has emerged as a potential therapeutic strategy. Although researchers have compared selected biofilm dispersion mechanisms,...

Amino acid reprogramming and biofilm-specific tricarboxylate transporters in PET-degrading Piscinibacter sakaiensis

ABSTRACT Plastic-degrading bacteria predominantly colonize polymer surfaces as biofilms, yet it remains unclear whether the biofilm phenotype contributes to metabolism beyond retaining extracellular enzymes. Here, we combine population-level RNA-sequencing across three conditions—biofilm cells on polyethylene terephtha...

Participation of the Transcriptional Regulator Fur in Modulating Biofilm Formation of the Salmonid Pathogen Yersinia ruckeri: Beyond Iron Homeostasis

Fur is identified as an important contributor to the normal progression toward mature and spatially organized Y. ruckeri biofilms and the Fur-dependent pathways and factors identified here provide candidates for future mechanistic studies and may ultimately help identify strategies to interfere with Y. ruckeri persiste...

Fructooligosaccharides Modulate Quorum-Sensing–Mediated Biofilm Formation in Lactiplantibacillus plantarum C8 via Amino Acid and Carbohydrate Metabolic Pathways

Oligosaccharides can support gut health by promoting the growth of beneficial bacteria like Lactobacillus, to help maintain gut balance and boost the immune system of the host. During the intestinal colonization, the forming biofilms can improve its adhesion and protect it from immune attacks and environmental stre...
