Confocal microscopy images of human adipose-derived mesenchymal stem cells exposed to palmitate: mitochondrial membrane potential (JC-1), mitochondrial superoxide (mtSOX Deep Red), mitochondrial network (MitoBright LT Green) and mitophagy (Mitophagy Dye)

Abstract

Original microscopy images supporting the article "Lipotoxic Palmitate Hyperpolarizes Mitochondria, Raises 8-oxo-dG Damage in Depleted mtDNA, and Converts Nuclear 5mC to 5hmC Genome-Wide in Human Adipose-Derived Mesenchymal Stem Cells" (International Journal of Molecular Sciences, submitted 2026). Human adipose-derived mesenchymal stem cells (one commercial lot) were imaged live on an Andor BC43 spinning-disk confocal microscope (40× objective) after 24 h without treatment (NT) or with 250 µM or 500 µM sodium palmitate, with a positive control for each assay. Probes: JC-1 (2 µmol/L; CCCP control), mtSOX Deep Red (10 µmol/L; Antimycin A control), MitoBright LT Green (0.1 µmol/L; CCCP control) and the Mitophagy Detection Kit (Mtphagy Dye 100 nmol/L, Lyso Dye 1 µmol/L; CCCP control); nuclei with Hoechst 33342. Version 1 contains the original images of the micrograph panels shown in Figures 2, 4 and 7 (c, d) of the article: for each panel, the snapshot exported from Imaris (PNG) and the deconvolved maximum-intensity projection of every channel of the same field (16-bit TIFF), which is the input of the image quantification. Original_Images_guide lists every file with its figure panel, condition and field of view. The quantification in the article uses all analysed fields (4–6 per condition). Later versions of this record will add the raw image files (Imaris .ims) of every analysed field, the 24 h time-lapse of the mitochondrial network (Figures 5–6) and the viability images (Figure 1).
