The Id2 induced block in T cell generation is overcome by TCR ab overexpression

TL;DR

It is shown that sustained Notch induced signals are required to seal the commitment of TCR ab /CD3 expressing cells into the TCR ab T cell lineage, which otherwise develop into cells phenotypically resembling NK cells or NKT cells.

p53 activation via pharmacologic MDM2 inhibition constrains exhaustion and enhances antitumor function of human CD8+ T cells

Adoptive T-cell therapies using chimeric antigen receptors (CARs) or engineered T-cell receptors (TCRs) have transformed cancer treatment but remain limited by poor persistence and progressive T-cell exhaustion. Manufacturing strategies that promote memory T-cell formation may improve therapeutic durability. Here we...

The Notch signaling pathway is a master regulator of CD8+ T cell exhaustion and differentiation during chronic infection

It is shown that the absence of Notch signal causes severe CD8+ T cell exhaustion during chronic LCMV infection and demonstrated that Notch signaling is essential to promote the differentiation of Tex progenitors into Tex effector-like cells.

Upregulation of CD90 correlates with T lineage commitment at the DN2a stage of T cell development

Early thymic progenitors (ETPs) retain the potential for alternative fates but commit to the T-cell lineage as they progress through the CD4−CD8− double-negative (DN) thymocyte stages early in T-cell development. Current evidence suggests that T-lineage commitment occurs during the transition from the DN2a to DN2b stag...

PRMT7 restricts CD8+ T cells expansion via the NF-ΚB pathway

Successful cellular immunotherapy for cancer utilising cytotoxic T lymphocytes (CTL) requires efficient expansion while maintaining effector function. Understanding how CTL expansion and function are regulated during the cell preparation process is thus important. Here, we show that T cell-specific deletion of Prmt7 us...

RNA-seq-based transcriptome analysis identifies TCR-expressing B1 cells as a multipotent lymphoid intermediate.

Classically, B and T lymphocytes are regarded as distinct lineages that can be readily distinguished by the receptors expressed on their surface. However, our recent study established that peritoneal B1 cells serve as precursors for extrathymic T cells through a TCR+CD19+ double-positive (DP) intermediate. To investiga...

Abstract IA005: Determinants of B cell fate and function in cancer

Effector B cell responses in solid malignancies are associated favorable response to immunotherapy. B cells can amplify anti-tumor immune responses via antibody production, antigen presentation, and pro-inflammatory cytokine release; yet B cells in cancer patients and tumor-bearing mice often fail to support these fu...
