Computer analyses supported its Keap1-targeting activity, identifying CS-11 as a promising Parkinson’s disease therapeutic candidate and showing the strongest neuroprotective effect in a Parkinson’s disease model.

Abstract

Oxidative stress and xenobiotic exposure disrupt redox homeostasis and contribute to neurodegenerative disease progression. The Keap1-Nrf2-ARE pathway is a key antioxidant defense mechanism, in which Nrf2 activates cytoprotective gene expression. Because disrupting the Keap1-Nrf2 interaction can enhance antioxidant responses, we assessed modified Cannabis sativa cannabinoids bearing C- and O-β-d-glucosides as potential Keap1 inhibitors. Building on our earlier fractionation and derivatization work, these compounds were evaluated for binding to the Keap1 BTB and KDC domains and for activation of Nrf2-mediated responses using in vitro and in silico methods. In a Parkinson’s disease model, CS-11 (4a) showed the strongest neuroprotective effect, with an EC50 of 100 nM. Treatment significantly increased cytoprotective gene expression, indicating enhanced Nrf2 transcriptional activity. CS-11 (4a) also restored tyrosine hydroxylase levels and protected dopaminergic neurons from cell death. Computational analyses supported its Keap1-targeting activity, identifying CS-11 (4a) as a promising Parkinson’s disease therapeutic candidate.

Betulin supplementation reduces oxidative stress in Alzheimer's disease via the activation of KEAP1/NRF2 pathway.

Betulin (BE) is extracted from pentacyclic triterpenoids of many natural plants and has antioxidation activities. However, the neuroprotective effects and underlying mechanism of Betulin in Alzheimer's disease (AD) have remained limited. Baicalein targets the Nrf2/Keap1 interaction and related stress pathways to mitigate drug-induced toxicity and reverse resistance in breast cancer: A review.

The cellular stress mediator Nrf2 and its negative regulator Keap1 are frequently dysregulated across breast cancer subtypes. As a master regulator of stress-related pathways, the Nrf2/Keap1 complex drives either cytoprotection or cell death, depending on the cellular context. Furthermore, its roles in promoting drug r...

The Discovery of GSK3227634: A Potent Inhaled Inhibitor of the KEAP1/NRF2 Protein–Protein Interaction for the Treatment of Chronic Obstructive Pulmonary Disease

KEAP1 is the key regulator of the NRF2-mediated cytoprotective response and a target for pathologies involving oxidative stress. Compounds that covalently modify KEAP1 to activate NRF2 are clinically validated; however, their chemical reactivity may drive increased off–target activity. A more selective approach involve...

Rational discovery of peptide inhibitors targeting Keap1: a focus on neurodegenerative diseases

Introduction The Kelch-like ECH-associated protein 1 (Keap1)/Nuclear factor erythroid 2-related factor 2 (Nrf2) pathway is a promising therapeutic target for neurodegenerative diseases (NDDs). Nrf2 activation mitigates key pathogenic mechanisms in NDDs by enhancing antioxidant defenses, suppressing neuroinflammation, i...
