Abstract

Zucchini yellow mosaic virus (ZYMV; genus Potyvirus), an aphid-transmitted pathogen, has become a major threat to melon (Cucumis melo) production in China, severely reducing both crop yield and fruit quality. Cross protection using attenuated virus strains is a key strategy for controlling plant viral diseases. However, few ZYMV vaccine candidates possess both multiple attenuating mutations and impaired aphid transmissibility. In this study, we identified that the conserved residues in helper component-protease (HC-Pro), glycine at position 322 (G322) and lysine at position 360 (K360), played essential roles in RNA silencing suppression and ZYMV virulence. To reduce the risk of mild strains mutating to virulent ones and eliminate aphid transmission of an attenuated ZYMV strain, we substituted G322 and K360 with alanine (A) and aspartic acid (D), respectively, and further replaced A in the conserved DAG motif of the coat protein (CP) with threonine (T), generating a triple-mutant designated ZYMV-ADT. ZYMV-ADT maintained genetic stability through three serial passages and caused infection without apparent mosaic symptoms at 45 days post-agroinfiltration. Moreover, ZYMV-ADT provided effective protection in melon plants against severe ZYMV infection with a 10-day protection period. This study provides an attenuated mutant out of a non-aphid transmissible ZYMV strain that offers a promising approach for managing ZYMV through cross protection.

Spray application of dsRNA reduces aphid transmitted ZYMV infection in Cucurbita pepo.

Exogenous application of double-stranded RNA (dsRNA) for protection against plant viruses has been demonstrated in a range of pathosystems. Here, we demonstrate the protective efficacy of spray-induced gene silencing (SIGS) in zucchini plants (Cucurbita pepo) against virulent zucchini yellow mosaic virus (ZYMV) upon me...

Molecular Characterization and Infectious Assay of Cymbidium Mosaic Virus in the Medicinal Plant Tetrastigma hemsleyanum in China

Tetrastigma hemsleyanum is a valuable and rare medicinal herb in the family Vitaceae . With its large‐scale cultivation in China, a virus‐like disease has emerged threatening both yield and quality in recent years. However, the viral pathogen has not been reported. Here, we detected a new isolate of Cymbidium mo...

Molecular and biological characterization of Peruvian isolates of zucchini yellow mosaic virus reveals differential severity and partial resistance breakdown in zucchini cultivars

Abstract Zucchini yellow mosaic virus (ZYMV) is one of the most important potyviruses affecting cucurbit crops worldwide and has recently been reported in Peru. However, information regarding the biological variability and host response of Peruvian isolates remains limited. This study aimed to characterize two Peruvian...

A novel pepper mild mottle virus-based vector for high-efficiency gene expression and silencing in pepper (Capsicum annuum)

Pepper ( Capsicum annuum ) is an economically important vegetable crop worldwide. However, progress in its functional genomics has been severely impeded by the low efficiency and time-consuming nature of stable genetic transformation. To address these limitations, plant virus-based vectors have been developed...

Base editing of EIF4E creates novel resistance alleles against bymoviruses in winter barley

The present study represents the first application of base editing in barley plants, using the EIF4E gene as an example, and two newly generated alleles led to resistance upon BaMMV inoculation without adverse effects on yield, proving this approach promising to generate new material for resistance breeding.
