A Bacterial Immune Memory Becomes a Genome Editor: A Historical Development Review of CRISPR-Cas Systems

Abstract

This article offers a historical development review of CRISPR-Cas systems, the adaptive immune architecture of bacteria and archaea that became, within a single decade, the most consequential genome editing technology in the history of biology. The review reconstructs the field's development across five phases: the observation of unexplained clustered repeats in the Escherichia coli genome in 1987 and the two decades of comparative genomics that named them and proposed, from the growing database of spacer sequences, that they recorded past viral exposures; the confirmation of the immune hypothesis in 2007, in an experiment performed not in a medical laboratory but in an industrial dairy cultures facility, where phage resistance was shown to be acquired, heritable, and spacer-sequence-specific; the mechanistic decade that established RNA-guided DNA cleavage, classified the cas gene families, and culminated in the 2012 demonstration that the Cas9 nuclease could be programmed with a single chimeric RNA to cut any chosen sequence; the editor era of 2013 and after, in which the system was deployed in human cells within weeks, diversified into base and prime editors, and moved through agriculture and medicine to the first approved CRISPR therapeutic; and the ethical rupture of the 2018 germline announcement, which confronted the field with governance questions its scientific predecessors had a generation to prepare for. Three synthetic claims are advanced. First, the technology's origin is a case study in the basic-applied symbiosis, since the function was proven in a yogurt factory and the editors were trained in molecular biology's curiosity-driven tier. Second, the hinge of the entire development was modularity, the separation of recognition from nuclease, which converted a defense system into a platform. Third, the field's ethics arrived on a different clock than its science, and the comparison with molecular biology's self-governance tradition exposes what was lost in the compression. An agenda is proposed spanning delivery, specificity assessment, and governance design.
