This study explored the molecular mechanisms underlying CRC immune evasion as well as the therapeutic effects of immune checkpoint inhibitors in microsatellite‐stable/proficient mismatch repair CRC.
Abstract
Colorectal cancer (CRC) is a major global health burden. While immune checkpoint inhibitors have greatly advanced cancer therapy, their therapeutic effects are unsatisfactory in microsatellite‐stable/proficient mismatch repair CRC. This study explored the molecular mechanisms underlying CRC immune evasion.
The objective response rate of immune checkpoint inhibitors in patients with gastric cancer is limited. Targeting specific subtypes of macrophages holds promise for expanding the potential population sensitive to immune checkpoint inhibitors. This study aims to evaluate the key role of TREM1+tumor‐associated macrophage...
Highlights • PCK1 is overexpressed in gastric cancer and promotes tumor growth and metastasis.• PCK1 activates the SRC1-JAK1-STAT3 cascade in gastric cancer cells.• The PCK1-SRC1-STAT3 pathway is a potential therapeutic target for gastric cancer.

Highlights • FJX1 is overexpressed in CRC and predicts poor patient survival.• FJX1 drives FOLFOX resistance by directly binding and activating PPARγ.• FJX1-PPARγ interaction promotes EMT, proliferation, and tumor migration.• Disrupting the FJX1-PPARγ axis with FX909 restores chemosensitivity.

The underlying mechanism of tumor microenvironment-induced dysfunction of conventional type 1 dendritic cells (cDC1) remains unclear. Zhang et al. identify transforming growth factor β-driven coactivator-associated arginine methyltransferase 1 (CARM1) as an epigenetic inhibitor of cDC1 cross-presentation. CARM1 is a ce...

Heterogeneity in cancer‐associated fibroblast (CAF) infiltration within the tumor microenvironment is closely associated with gastric cancer progression and immune evasion; however, precise CAF‐related diagnostic markers and actionable therapeutic targets remain scarce.
