It is reported that STAT3 inhibition eliminates multiple myeloma stem-like cells while eliciting hallmarks of immunogenic cell death, yet in vivo validation remains necessary before this strategy can be considered immunogenic.
Abstract
ABSTRACT In a recent paper published in Blood Science, Sha et al. report that STAT3 inhibition eliminates multiple myeloma stem-like cells while eliciting hallmarks of immunogenic cell death. These highly intriguing findings connect stemness, integrated stress response, and antitumor immunity, yet in vivo validation remains necessary before this strategy can be considered immunogenic.
In this issue of Cell, Lei et al. reveal a reciprocal circuit in which cuproptotic tumor cells promote dendritic cell activation and CD8+ T cell priming, whereas T cell-derived IFN-γ sensitizes tumor cells to FDX1-dependent cuproptosis. This self-reinforcing interaction provides a mechanistic rationale for combining cu...
This study provides evidence for a key role of TNFR1, as expressed by malignant cells, in orchestrating a pro-tumor immunosuppressive environment through secondary inflammatory mediators. Results highlight the relevance of interfering TNFα/TNFR1 in cancer immunotherapy.

In this issue of Cancer Cell, Kehl et al. construct a single-cell atlas of bone marrow T cells from patients with bone marrow-resident malignancies, identifying tumor-reactive populations existing in a state of latent competence. A 15-gene signature identifies this subset, which is expanded by immunotherapies and predi...

In a recent paper published in Nature, Yuan et al. described an acid-responsive membranolytic peptide that temporally separates lysosomes from later plasma membrane rupture, thereby converting oncolysis into immunogenic cell death. This elegant design permits systemic administration, elicits antigen-specific immunity,...
