TL;DR

Intacranial TX103 infusion demonstrated acceptable safety and encouraging efficacy in rGBM, supporting a future phase 2 evaluation at the RP2D, and Cerebrospinal fluid showed a marked increase in CAR copy numbers and cytokine release, with minimal peripheral activity and no cumulative toxicity after repeated infusions.

Autologous Glypican-3-Targeted, Armored CAR T Cells in Patients with Advanced Solid Tumors: A Phase 1 Dose-Escalation Study of TAK-102.

Findings support a manageable safety profile for TAK-102 and provide early evidence of biological activity in solid tumors, and are warranted to optimize dosing, improve CAR T-cell persistence, and evaluate efficacy in larger patient cohorts.

Sequencing BCMA-Directed Therapies in Relapsed/Refractory Multiple Myeloma: A Real-World Comparison of CAR T-Cell Therapy and Bispecific Antibodies

Simple Summary B-cell maturation antigen (BCMA)-directed therapies (BDT) are approved for the treatment of relapsed/refractory multiple myeloma (RRMM). Several treatment modalities exist including chimeric antigen receptor T-cell therapy (CAR-T), bispecific antibodies (BsAbs), and antibody-drug conjugates (ADC). The ef...

MK-4002, a B-cell maturation antigen-targeting T-cell engager, in relapsed or refractory multiple myeloma: a first-in-human, open-label, phase 1 study.

BACKGROUND The optimal treatment strategy for relapsed or refractory multiple myeloma remains unclear. This first-in-human study aimed to evaluate the safety, preliminary activity, and pharmacokinetics of the trispecific B-cell maturation antigen-targeting T-cell engager MK-4002 in relapsed or refractory multiple myelo...

Safety and Biomarker Analysis of NRG-LU004: Phase I Trial of Accelerated or Conventionally Fractionated Radiotherapy Combined With Durvalumab in PD-L1 High Locally Advanced Non-Small Cell Lung Cancer

Background. Patients with advanced non-small cell lung cancer (NSCLC) and high PD-L1 (>50%) expression show improved outcomes with immune checkpoint inhibitors compared to chemotherapy. We hypothesized that concurrent durvalumab and radiotherapy (RT) without concurrent chemotherapy would be safe and feasible in patient...

Tocilizumab, atezolizumab, and stereotactic radiotherapy in recurrent glioblastoma: primary analysis of safety and efficacy of a phase 2 trial, NRG Oncology-BN010.

The findings suggest that the combination of tocilizumab, atezolizumab, and FSRT was safe and well tolerated in patients with rGBM but did not achieve the pre-specified efficacy outcome.
