TL;DR

A CRISPR/Cas9-based single-cell lineage tracing system with paired multiomics profiling in human pluripotent stem cell-derived neuromesodermal organoids to reconstruct high-resolution lineage relationships spanning 50 days of differentiation provides a clonal-resolution lineage map of human NMP differentiation and advances understanding of human posterior trunk development.

Brachyury expression levels predict lineage potential and axis-forming ability of in vitro-derived neuromesodermal progenitors

This dual Sox2/Tbxt double reporter cell line provides support for unsuspected heterogeneity in NMPs, together with evidence for a role of these transcription factors in directing cell fate to drive axis elongation.

Hox/Meis-dependent gene-regulatory transition underlies cardiopharyngeal neural crest diversification

Neural crest cells (NCCs) are multipotent migratory cells essential for cardiac development, yet the lineage trajectories and gene regulatory networks underlying their differentiation in the cardiopharyngeal region remain unclear. Here, we integrate single-cell RNA-seq, spatial transcriptomics, and multiomic analyses t...

Transcriptomic Analysis Identifies Transient Mesendodermal State and Lineage Divergence in Human Pluripotent Stem Cell Differentiation

These findings demonstrate that comparative transcriptomics can resolve developmental intermediates and cellular-state dynamics during human germ layer specification, providing a framework for evaluating lineage commitment beyond endpoint canonical marker expression, and to inform strategies for optimizing or redirecti...

In utero barcoding of mouse mesoderm reveals the clonal architecture of organ mesenchyme

The mesoderm coordinates organogenesis and generates the cardiovascular, visceral and musculoskeletal systems, yet in mammals this germ layer is difficult to access, impeding its genetic manipulation and study. Here, we establish embryonic day (E) 7.5 exocoelomic cavity nano-injection as a scalable route for selective...
