A multimodal model for clinically defined MCI integrating plasma biomarkers and brain microstructural features: a dual-cohort external validation study
A multimodal signature combining two selected DTI features with three prespecified plasma biomarkers and demographic covariates showed reproducible discrimination of clinically defined MCI across independent cohorts, and support further evaluation of this approach as an adjunctive MCI risk-stratification strategy.
Background Identifying candidate biomarkers for preclinical dementia could improve our understanding of underlying biological pathways leading to vascular brain injury (VBI) and neurodegeneration. Objective To identify plasma protein markers of MRI markers of VBI and amyloid- and tau-PET burden in cognitively healthy a...

An explainable multimodal machine-learning framework to predict progression from MCI to dementia within a clinically meaningful two-year period is developed and the convergence of cognitive impairment and region-specific neurodegeneration emerged as the strongest indicator of conversion risk.

Bomarker-confirmed AD comprised internally reproducible atrophy trajectories with distinct cognitive, vascular, and molecular profiles and may complement fluid biomarkers and conventional volumetry for patient stratification and prognosis.

Quantitative T1 mapping (qT1) is a magnetic resonance imaging (MRI) biomarker of brain microstructural changes; however, its application in Alzheimer disease (AD) remains limited. We compared cortico-limbic qT1 values between patients with AD and healthy controls (HCs) and examined their relationship with amyloid burde...

The iSCN approach identified reliable patterns of topological and connectivity impairment across MS and its clinical stratifications, providing insight into MS neuropathology and guiding future diagnostic and therapeutic biomarker development.
INTRODUCTION White matter (WM) functional activity, quantified as slow-4 fractional amplitude of low-frequency fluctuations (fALFF; 0.027–0.073 Hz), may reflect Alzheimer’s disease (AD) pathology, but its utility relative to established structural imaging markers remains poorly characterized. METHODS We analyzed 369 AD...
