Abstract

Hepatocellular carcinoma (HCC) remains a formidable malignancy with limited response to conventional monotherapies, largely due to its complicated pathogenesis and useful cellular defense systems. In this study, we constructed a dual-targeting polymeric micelle (S&A@TPD/HG PMs), co-delivering sorafenib (SOR), and artesunate (ART) to achieve synergistic ferroptosis-based therapy against HCC. The micelle features a hyaluronic acid (HA) shell functionalized with glycyrrhetinic acid (GA) for active dual targeting to CD44 and GA receptors overexpressed on HCC cells; while its triphenylphosphonium (TPP)-decorated inner core facilitates mitochondrial anchoring and co-conveys the drugs, enabling stepwise subcellular targeting. Compared with free drugs or single-drug-loaded micelles, S&A@TPD/HG PMs effectively induced mitochondrial damage by depleting glutathione (GSH), and downregulating GSH peroxidase 4 (GPX4) expression to disrupt the mitochondrial antioxidant defense system, elevating intracellular Fe2+ to increase reactive oxygen species (ROS) and lipid peroxidation (LPO) production, thereby compromising mitochondrial redox homeostasis and potentiating ferroptosis jointly. This ferroptosis-inducing activity yielded potent in vivo antitumor efficacy in Huh-7 tumor-bearing nude mice, with a tumor inhibition rate of 74.23% and minimal systemic toxicity. Collectively, the organelle-specific targeting combined with co-delivery of synergistic drug pairs represents a promising strategy for combating malignancies and provides a blueprint for the rational design of multifunctional nanosystems.

Dual-Targeted Galactose-TPP Dendrimers Modulate Mitochondrial Function and Downstream Outcomes in Hepatocellular Carcinoma

Hepatocellular carcinoma (HCC) remains therapeutically challenging. Sorafenib, the standard first-line therapy for advanced HCC, suffers from poor aqueous solubility, non-specific biodistribution, dose-limiting toxicities, and the emergence of mitochondrial-mediated resistance. Here, we engineered a dual-targeted den...

Thermosensitive RGD-modified liposomes co-loading Fe3O4 and sorafenib for synergistic photothermal-chemotherapy of hepatocellular carcinoma.

Sorafenib (SRF) therapy for hepatocellular carcinoma (HCC) is limited by dose-limiting toxicities, inadequate tumor exposure, and the inability to confine drug action spatiotemporally, leading to suboptimal efficacy and a narrow therapeutic window. Here we report an RGD-functionalized DPPC-based thermosensitive liposom...

All-in-One Mitochondria-Targeted Iron-Based MOF “Nanomissiles” for Synergistically Potentiated Tumor Ferroptosis, Dual Phototherapy, and Chemotherapy

Pronounced heterogeneity and adaptability of tumors inherently curtail monotherapeutic efficacy, necessitating rationally integrated multimodal regimens that enable precise intervention, overcome resistance, and eradicate heterogeneous cell populations within the complex tumor microenvironment (TME). All-in-one mitoc...

Supramolecular self-assembled nanomedicine targeting fatty acid metabolism to potentiate ferroptosis-enhanced photoimmunotherapy in triple-negative breast cancer

Triple-negative breast cancer (TNBC) presents a serious therapeutic challenge, largely owing to its metabolic adaptability and the immunosuppressive nature of its tumor microenvironment (TME). Photodynamic therapy (PDT) can cause oxidative injury and induce immunogenic cell death (ICD); however, its antitumor activity...

Ursolic acid sensitizes hepatoma cells to sulfasalazine-induced ferroptosis via AMPK/BECN1/SLC7A11-mediated redox disruption in vitro and in vivo

Targeting ferroptosis has emerged as a promising anti-cancer strategy. A Food and Drug Administration (FDA)-approved drug, sulfasalazine (SAS), also a ferroptosis inducer, is being explored as a potential treatment drug for cancer. However, long-term high-dose SAS treatment may induce several adverse effects in the...
