The optimization of PEGylation reaction yield is critical for the efficient production of therapeutic bioconjugates. This study systematically investigates the influence of three distinct classes of additives -organic cosolvents, nonreactive poly(ethylene glycols) (PEGs) of varying molecular weights, and surfactants- on the conjugation efficiency of a model protein. Quantitative analysis via SDS-PAGE densitometry revealed that the addition of 40% (v/v) DMSO significantly enhanced the reaction yield compared to the aqueous control, while methanol resulted in a marked decrease in efficiency. These results were rationalized using Hansen solubility parameters, which indicated that the high dispersion cohesion (18.4 MPa1/2) of DMSO promotes superior solvation of PEG chains, thereby minimizing competitive hydrolysis pathways. Furthermore, the inclusion of nonfunctionalized PEGs (4 and 8 kDa) provided a marginal increase in conjugation, likely due to the excluded volume effect. In contrast, the anionic surfactant SDS severely inhibited the reaction, a phenomenon attributed to electrostatic repulsion between the charged SDS-PEG-protein complexes. The nonionic surfactant polysorbate 20 exhibited a neutral effect on yield, suggesting its utility primarily as a protein stabilizer without compromising conjugation. These findings provide a comprehensive framework for the strategic selection of additives to modulate the physicochemical environment and maximize PEGylation efficiency.
Abstract This paper focuses on the development of a sterilizing gel of 4-chloro-3,5-dimethylphenol (CDMP) arising from the presence of wormlike micelles, as detected by dynamic viscosity measurements. Surprisingly, the gel state was observed not only in the ternary system consisting of the anionic surfactant sodium dod...

In various biomedical applications, crosslinking of biomaterial networks with high spatiotemporal control is of key importance. In this context, diazirine photo-crosslinkers, like trifluoromethyl aryl diazirines, have evolved as radical-free, highly-efficient go-to agents. However, aryl derivatives face solubility issu...

Abstract Using cardanol-based nonionic surfactant, cardanol polyoxyethylene ether (NSF) with an EO addition number of 7 as the precursor, the anionic-nonionic surfactant, cardanol polyoxyethylene sulfate (NES) was prepared via sulfonation with chlorosulfonic acid. The molecular structures were characterized by FT-IR, a...
Background: Stable aqueous alumina suspensions depend strongly on the molecular architecture and relative proportion of adsorbing dispersants. Methods: Two polyethylene-glycol (PEG) esterification-product families were investigated: an acrylic-acid/PEG product (X) and a citric-acid/PEG product (Y).Results: Orthogonal sc...

A series of low-molecular-weight chitosan derivatives with varying degrees of hydrophobic and cationic substitution was synthesized using a reductive alkylation method. At the first stage, a hydrophobic C14-alkyloxybenzyl moiety was introduced into the macromolecule; at the second stage, a cationic quaternary ammonium...
