These findings identify distinct but concurrent SRSF1-regulated programs: transcriptional activation of genes associated with cellular signaling and organization and alternative splicing modulating genome-maintenance and p53-regulatory genes.

Abstract

Serine/arginine-rich splicing factor 1 (SRSF1) regulates alternative splicing and acts as a proto-oncogene, but its role in transcription remains poorly defined. We combined time-course RNA-seq with experimental validation to define SRSF1-dependent transcriptional and splicing programs. We validated 67 of 143 alternative-splicing events selected for experimental analysis. Among these, 44 were not annotated in GENCODE, including 15 containing previously unannotated exons. Thirty-five events showed concordant regulation in HEK293 and breast and prostate cancer cell lines, and 17 events in 16 genes affected p53 regulation or p53-dependent functions. SRSF1 overexpression reduced total p53, increased basal Ser15 phosphorylation, and increased apoptosis under basal and cisplatin-treated conditions but not following doxorubicin treatment. SRSF1 also increased transcription of 28 genes, 18 of which showed concordant regulation across the same cellular models. Mechanistic studies linked SRSF1-dependent transcriptional activation to P-TEFb mobilization from the 7SK snRNP, which controls P-TEFb availability for RNA polymerase II elongation. These findings identify distinct but concurrent SRSF1-regulated programs: transcriptional activation of genes associated with cellular signaling and organization and alternative splicing modulating genome-maintenance and p53-regulatory genes.

De novo variants in the essential splicing factor gene SRSF3 cause variable developmental phenotypes.

SRSF3 (SRp20), a member of the serine/arginine-rich protein family of splicing factors, plays a central role in constitutive and alternative splicing. The C-terminal arginine/serine-rich domain of SRSF3 undergoes dynamic phosphorylation cycles that regulate its subcellular localization, protein-protein interactions, an...

Mechanistic insight into SP1-mediated transcriptional activation of the long non-coding RNA MIR100HG.

Findings provide the first direct evidence that SP1 transcriptionally activates MIR100HG while simultaneously suppressing malignant cellular phenotypes, revealing a novel SP1-MIR100HG regulatory axis that may contribute to hepatocellular carcinoma progression in an oxygen-dependent manner.

RBM39 regulates malignant progression in lung adenocarcinoma cells through alternative splicing of NRG1-206

To investigate the function of RBM39 in lung adenocarcinoma (LUAD), with a particular focus on its regulatory effect on the alternative splicing of NRG1-206 and its role in tumor progression. Differential expression analysis of RNA binding proteins (RBPs) in LUAD samples was performed using TCGA datasets. RBM39 exp...

Intron Retention: Organisation, Genome and Biological Roles and Associated Diseases

Alternative splicing expands transcript and protein diversity by generating multiple isoforms from individual genes. Intron retention (IR), a major form of alternative splicing, occurs when intronic sequences remain in mature RNA transcripts and is now recognized as a regulated mechanism of post-transcriptional gene co...
