Abstract

Simple Summary Breast cancer remains a leading cause of cancer-related death among women, and cisplatin resistance limits chemotherapy efficacy in patients with advanced breast cancer. Here, we investigated the role of ARMCX3 in breast cancer progression and cisplatin responsiveness. ARMCX3 depletion reduced cell growth and suppressed migratory and invasive capacities while increasing sensitivity to cisplatin treatment in vitro, although re-expression of ARMCX3 rescued these phenotypes. Transcriptomic and molecular analyses further suggested that ARMCX3 depletion was associated with increased p53-related signaling, altered DNA damage response signaling and enhanced apoptosis following cisplatin treatment. Overall, our findings provide further insight into the role of ARMCX3 in breast cancer progression and cisplatin responsiveness and support further investigation of ARMCX3-associated signaling in the context of cisplatin response.
