P19 represents a promising PIM1-potent, PKC-sparing multi-kinase lead for further TNBC optimization and reduced PIM1-GBP1 association and GBP1 phosphorylation, suggesting that the PIM1/GBP1 axis may contribute to its cellular activity.
Abstract
PIM1 has emerged as a compelling target for triple-negative breast cancer (TNBC), which lacks effective treatments. Through integrated virtual screening and in vitro validation, N6 was identified as a potent PIM1-active indolocarbazole lead, although its strong PKC inhibition indicated a limited selectivity window. Structural comparison of PIM1 and PKC ATP-binding pockets revealed differences, including the Phe644 region in PKCη, that could be exploited to reduce PKC binding. A deep-learning/genetic-algorithm workflow yielded P19, which retained potent PIM1 inhibition (IC50 = 3.6 nM) while showing lower cytotoxicity toward MCF-10A cells and reduced inhibition of PKCδ/ε/η relative to N6. In an orthotopic TNBC mouse model, P19 (25 mg/kg) achieved potent tumor growth inhibition (TGI = 66.3%). Mechanistically, P19 reduced PIM1-GBP1 association and GBP1 phosphorylation, suggesting that the PIM1/GBP1 axis may contribute to its cellular activity. Overall, P19 represents a promising PIM1-potent, PKC-sparing multi-kinase lead for further TNBC optimization.
PKMYT1 mediates the inhibitory phosphorylation of CDK1 and represents a promising synthetic lethal target in p53-deficient or CCNE1-amplified malignancies. A series of PKMYT1 inhibitors featuring a novel monocyclic pyridine core structure was discovered, exhibiting improved selectivity and enhanced drug-like properties...
The design, synthesis, and biological evaluation of a series of novel PAK1 inhibitors guided by a conformation-driven optimization strategy establish ZMF-28 as a promising therapeutic candidate for TNBC and highlight the value of conformation-guided design in kinase inhibitor discovery.

Inhibiting PI3Kα has demonstrated promising therapeutic effects for patients with HR+/HER2- and PIK3CA-mutated advanced or metastatic breast cancer. However, hyperglycemia and other notable adverse events (AE) associated with concomitant wild-type inhibition remain a challenge and limit dosing. Selectively targeting th...

Abstract Pancreatic ductal adenocarcinoma (PDAC) remains a lethal malignancy with limited targeted therapeutic options. Mitogen-activated protein kinase kinase kinase kinase 4 (MAP4K4) has been implicated in PDAC progression, but the development of effective MAP4K4 inhibitors requires further improvement in potency, se...

PKMYT1 is a critical regulator of the cell cycle and has emerged as a promising synthetic lethal target for CCNE1-amplified cancers. In this study, we report the design, synthesis, and biological evaluation of a novel series of 4-aminoquinazoline-based PKMYT1 inhibitors. A hydrogen bond donor-to-acceptor conversion str...
