Findings reveal that FTO and IGF2BP1 co-regulate the differentiation of intramuscular adipocytes through post-transcriptional modification of BCL11A, providing novel insights into m6A-mediated lipid deposition in goat muscle.

Abstract

Intramuscular fat (IMF) plays a critical role in determining the flavor of goat meat. Studies have shown that N6-methyladenosine (m6A) modification of mRNA exerts a key influence on adipocyte differentiation and lipid metabolism. However, whether m6A−modified mRNA regulates the differentiation of intramuscular adipocytes in goats remains unclear. This study constructed the m6A methylation landscape during the differentiation of goat intramuscular adipocytes using MeRIP-seq and RNA-seq, and identified BCL11A as a key regulator exhibiting synchronized changes in both m6A modification and expression. Functional assays demonstrated that BCL11A promotes lipid accumulation and upregulates adipogenic markers ( C/EBPα , PPARγ , SREBP1 , FASN ). Mechanistically, the demethylase FTO directly targets the m6A site on BCL11A , while the reader protein IGF2BP1 recognizes this modification and reduces BCL11A mRNA stability. FTO promotes adipogenesis, whereas knockdown of IGF2BP1 shortens the half-life of BCL11A and inhibits differentiation. These findings reveal that FTO and IGF2BP1 co-regulate the differentiation of intramuscular adipocytes through post-transcriptional modification of BCL11A , providing novel insights into m6A-mediated lipid deposition in goat muscle.

Aloe-Emodin Modifies Adipogenic Genes and the Expression of miR-143 and miR-155 in 3T3-L1 Cells

Aloe-emodin can upregulate transcription factors (PPARγ and C/EBPβ) as well as miR-143 and miR-155, suggesting its potential as a modulator of adipogenic gene or miRNA expression in this exploratory setting.

Cinnamaldehyde combined with 18β-glycyrrhetinic acid promotes the differentiation of C3H10T1/2 MSCs into thermogenic adipocytes through PGC1α/FNDC5/UCP1 pathway

ABSTRACT Finding bioactive substances that can promote the de novo differentiation of stem cells into thermogenic adipocytes is a promising anti-obesity strategy. Cinnamaldehyde (CM) and 18β-glycyrrhetinic acid (GA) are the main active ingredients in Chinese herbal medicine. In this study, C3H10T1/2 mesenchymal stem ce...

Study on the Role of FHL2 in Chicken: Promoting Bone Growth and Inhibiting Fat Formation via Regulating BMSC Differentiation Balance

Simple Summary In some chickens, high body weight is accompanied by a low dressing percentage because growth is preferentially directed to bone and fat rather than muscle. Our study identified four and a half LIM domains protein 2 (FHL2) as a key gene that promoted osteogenic differentiation and suppressed adipogenic d...

Emodin alleviates endometrial fibrosis via METTL3/YTHDF2-mediated m6A modification of PPARα and the fatty acid oxidation pathway

This study aimed to investigate the anti-fibrotic mechanism of emodin in endometrial stromal cells (ESCs), focusing on the N6-methyladenosine (m6A) modification of PPARα. An in vitro fibrosis model was established by treating ESCs with TGF-β1. Emodin was then administered to the cells, and its effects on fibrosis, ce...
