TL;DR

Clinical benefit was greatest when venetoclax was administered earlier, preceding the emergence of transcriptomic reprogramming that reduces BCL2 dependency, and findings support transcriptomic biomarker guided, subtype specific venetoclax based treatment strategies to optimize outcomes in patients with t(11;14) MM.

Belantamab mafodotin plus venetoclax in t(11;14)-positive relapsed or refractory multiple myeloma – the BELI(E)VE trial

The BELI(E)VE trial aims to establish a novel personalized therapeutic approach for a specific subset of MM patients by combining belantamab mafodotin and venetoclax, potentially inducing deep and durable remissions in t(11;14)-positive RRMM patients.

Genomic and clinical determinants of response to Azacitidine plus venetoclax in acute myeloid leukemia: Results from the HM-SCREEN-Japan 02 study

The combination of azacitidine and venetoclax (Aza/Ven) is an effective treatment for acute myeloid leukemia (AML); however, biological factors underlying heterogeneous treatment responses across diverse genetic backgrounds remain incompletely defined. We analyzed 97 AML patients treated with Aza/Ven in the prospective...

BCMA Targeted Therapies and Their Impact on Outcomes in Relapsed or Refractory Multiple Myeloma: Results From the French EMMY Cohort.

OBJECTIVES B-cell maturation antigen (BCMA)-targeted therapies have recently transformed the management of relapsed or refractory multiple myeloma (RRMM). This study aimed to evaluate their implementation and real-world impact within EMMY, a French longitudinal cohort. METHODS Patients initiating a fourth or later li...

Therapeutic strategies in the post-venetoclax era of acute myeloid leukemia

The therapeutic landscape of acute myeloid leukemia (AML) has changed substantially over the past decade with the incorporation of molecularly targeted agents, lower-intensity venetoclax-based regimens, and increasingly refined genetic risk stratification. Among these advances, the combination of venetoclax with hypome...

Molecular predictors of minimal residual disease in newly diagnosed multiple myeloma treated with quadruplet therapy.

Quadruplet therapy, combining anti-CD38 antibodies, immunomodulatory drugs, proteasome inhibitors and dexamethasone, is now the standard induction treatment for newly diagnosed multiple myeloma (NDMM). However, treatment response is heterogeneous and molecular predictors are needed to guide early therapeutic decisions....

TP53-Altered Aggressive Large B-Cell Lymphoma (LBCL) Outcomes by Treatment Modality: A Multicenter Cohort From 14 US Centers.

Advances in the treatment of large B-cell lymphoma (LBCL) have expanded therapeutic options beyond conventional chemoimmunotherapy to include cellular and targeted therapies. There is growing interest in identifying subsets of LBCL that may benefit from these novel approaches based on histopathologic and molecular feat...
