EPG5 is position EPG5 as a novel orchestrator of iron compartmentalization and the role of TBK1-NCOA4/TAX1BP1 axis in iron homeostasis and related stress response is highlighted.
Abstract
Intracellular iron is distributed across functionally distinct pools though the mechanisms regulating its compartmentalization remain unclear. Here we show that EPG5, a tethering factor required for autophagosome-lysosome fusion, governs iron compartmentalization through dual mechanisms: direct involvement in autophagy and indirect regulation of ferritinophagy initiation. A systematic comparison of autophagy genes indicates that EPG5 deficiency uniquely decreases hemosiderin and increases the labile iron pool, a profile distinct from ATG5, ATG14, FIP200, or WDR45 deficiencies. Mechanistically, EPG5 loss disrupts autophagosome maturation and endocytic trafficking, causing ferritin condensates and iron transporters to accumulate in aberrant autophagosomes and endosomes. This blockade triggers compensatory TBK1 activation. TBK1 subsequently phosphorylates TAX1BP1 and interacts with the ferritinophagy receptors NCOA4, inhibiting receptor homo-oligomerization and modulating ferritinophagic flux. These findings position EPG5 as a novel orchestrator of iron compartmentalization and highlight the role of TBK1-NCOA4/TAX1BP1 axis in iron homeostasis and related stress response.
Iron homeostasis is a central component of host defense responses. In mammals, iron availability is rapidly adjusted through receptor mediated pathways, whereas it remains unclear how plants regulate iron homeostasis during the earliest stages of immune activation. Here, we identify a rapid iron regulatory mechanism ce...

Ferritinophagy links molecular recognition of ferritin to the redistribution of redox-active iron across intracellular compartments. This critical narrative review examines nuclear receptor coactivator 4 (NCOA4)-mediated ferritinophagy as a multiscale process in which NCOA4-FTH1 binding and ferritin condensation govern...

Abstract Sequestosome 1 (SQSTM1 or p62) is a multi-domain protein that functions as a scaffold for aggregating proteins into cytoplasmic inclusions and mediating their turnover via selective autophagy. However, p62 is more than an autophagy scaffold or adaptor protein; it’s linked with endo/exosomal protein trafficking...

GlRac is established as a central regulator of an ATG8-independent autophagy-like pathway in Giardia regulated by GlRac, the parasite's sole Rho family GTPase, and demonstrates that this parasite retains key structural, regulatory, and degradation-associated features of autophagy despite the apparent absence of most ca...

At the center of iron homeostasis, macrophages play a crucial role by phagocytosing senescent erythrocytes while nursing erythrocyte progenitors. A major route of cellular iron acquisition occurs via transferrin receptor 1 (TFR1)-mediated endocytosis of transferrin-bound iron, a process that critically depends on endos...
