TL;DR

A novel PET tracer, [18F]XW-032, was developed via ruthenium-mediated deoxyfluorination and effectively visualized regional IDO1 heterogeneity in the AD brain, positioning it as a potent molecular tool for early AD diagnosis and for monitoring the efficacy of IDO1-targeted therapeutic interventions.

Abstract

IDO1-mediated dysregulation of the kynurenine pathway is a pivotal driver of neuroinflammation in Alzheimer's disease (AD). Despite its clinical significance, PET probes specifically targeting IDO1 in the AD brain remain scarce. In this study, we first established a strong correlation between IDO1 expression and AD progression through bioinformatic analysis. Subsequently, a novel PET tracer, [18F]XW-032, was developed via ruthenium-mediated deoxyfluorination. [18F]XW-032 demonstrated superior blood-brain barrier (BBB) permeability and a favorable metabolic profile. In vitro autoradiography and in vivo dynamic PET/CT imaging in 5XFAD mice confirmed its high binding specificity and robust imaging performance, further validated by competitive blocking studies with unlabeled IDO1 inhibitors. Notably, [18F]XW-032 effectively visualized regional IDO1 heterogeneity in the AD brain. These findings position [18F]XW-032 as a potent molecular tool for early AD diagnosis and for monitoring the efficacy of IDO1-targeted therapeutic interventions.

Synthesis and Preclinical Evaluation of 18F-CB86, a Novel Imidazole[1,2-a] Pyridine-Based TSPO Tracer

Background: The translocator protein (TSPO), an 18 kDa protein localized to the outer mitochondrial membrane, is a recognized biomarker of neuroinflammation. However, its clinical translation has been limited by factors such as poor specificity, low signal-to-noise ratio, and the influence of genetic polymorphisms (rs6...

Current Landscape of Glycogen Synthase Kinase-3β PET Tracers: Progress, Limitations, and Perspectives—A Narrative Review

Background/Objectives: Glycogen synthase kinase-3β (GSK-3β) serves as a crucial molecular target for Alzheimer’s disease and other neurological disorders. Positron emission tomography (PET) enables non-invasive quantification of GSK-3β in vivo, offering value for early diagnosis and mechanistic studies. However, despit...

Design, Synthesis and Evaluation Donepezil-Based Dual BuChE/FAAH Inhibitors for Alzheimer’s Disease Therapy

Alzheimer’s disease (AD) is multifactorial, prompting multi-target therapy. A series of donepezil-based derivatives were designed, synthesized, and evaluated as dual inhibitors of human butyrylcholinesterase (hBuChE) and fatty acid amide hydrolase (hFAAH). Among them, lead G15 displayed potent and balanced inhibition...

First in vivo evaluation of the radiolabelled Hsp90 inhibitor [11C]Onalespib for cancer and brain PET imaging

Background: Heat shock protein 90 (Hsp90) is a key molecular chaperone involved in maintaining proteostasis, and its function and aberrant expression are linked to tumour progression and to neurodegeneration. Its overexpression in cancers and the clinical development of Hsp90 inhibitors, including the recently approved...

Methods for assessing binding of a small-molecule radioligand to soluble enzyme isocitrate dehydrogenase 1 variants

Isocitrate dehydrogenase 1 (IDH1) is a cytosolic NADP+-dependent enzyme involved in cellular redox homeostasis and 2-oxoglutarate (2-OG) metabolism. Glioma-associated mutations in IDH1 confer a neomorphic enzymatic activity, resulting in the production and accumulation of the oncometabolite D-2-hydroxyglutarate (2-HG),...
