Abstract

To explore the total flavonoids extracted from Periploca forrestii (TFPF) ’s potential benefits in managing ulcerative colitis (UC) , an animal experiment was conducted. Mice were divided into a normal control group, a dextran sulfate sodium (DSS)-induced UC model group, a positive drug control group (given sulfasalazine at 200 mg/kg), and experimental groups treated with different TFPF dosages (25, 50, 100 mg/kg). The UC model was established by giving mice free access to 3% (w/v) DSS-supplemented drinking water for 7 days. During the modeling period, the experimental groups received daily oral sulfasalazine or TFPF via gavage, while the control and model groups were given the same volume of vehicle. After the intervention, parameters like Disease Activity Index (DAI), colon length, serum inflammatory cytokine levels, and colonic histopathology were evaluated. Results showed that TFPF, especially at 100 mg/kg, effectively reduced elevated DAI, prevented colon shortening, lowered serum cytokine concentrations, and improved colonic mucosal integrity. High-dose TFPF had similar therapeutic efficacy to sulfasalazine. These findings indicate that TFPF, especially in large quantities, has remarkable potential in alleviating DSS-triggered UC in mice, paving the way for further research and development of Periploca forrestii as a treatment option.
