Neoadjuvant cetuximab plus tislelizumab plus tislelizumab showed feasible delivery, encouraging antitumor activity, and acceptable safety in patients with locally advanced oral squamous cell carcinoma who were medically unsuitable for conventional cytotoxic chemotherapy.
Abstract
Background
Chemotherapy-free neoadjuvant strategies remain undefined for patients with locally advanced oral squamous cell carcinoma (OSCC) who are medically unsuitable for conventional cytotoxic chemotherapy.
Methods
In this prospective single-arm pilot trial, 15 patients received two cycles of neoadjuvant tislelizumab plus cetuximab before planned surgery. The primary endpoint was major pathological response (MPR).
Results
All patients completed neoadjuvant treatment and underwent radiographic assessment. The objective response rate was 73.3%. Twelve patients underwent surgery; five achieved MPR and two achieved pathological complete response. At a median follow-up of 23 months, the 2-year event-free survival and overall survival rates were 66.0% and 78.3%, respectively. No treatment-related deaths occurred.
Conclusions
Neoadjuvant cetuximab plus tislelizumab showed feasible delivery, encouraging antitumor activity, and acceptable safety in patients with locally advanced OSCC who were medically unsuitable for conventional cytotoxic chemotherapy.
Neoadjuvant chemoimmunotherapy is increasingly being explored for resectable, locally advanced oral squamous cell carcinoma, and yet real-world evidence regarding its effectiveness and safety remains limited. In addition, a proportion of patients do not proceed to surgery after neoadjuvant treatment, and outcomes in th...

BACKGROUND
Neoadjuvant therapy can improve the survival outcomes of patients with borderline resectable pancreatic cancer (BRPC). However, standard neoadjuvant regimens remain lacking. This study evaluated the clinical feasibility of tislelizumab plus gemcitabine and nab-paclitaxel in the neoadjuvant setting for BRPC....

Neoadjuvant chemo-immunotherapy (NACI) demonstrates promising efficacy in locally advanced head and neck squamous cell carcinoma (LA-HNSCC), yet its effect on postoperative radiotherapy/chemoradiotherapy (PORT/POCRT) remains undefined. This open-label phase 2 study (NCT05476965) assessed NACI efficacy and its role in g...

BACKGROUND
Technically resectable locally advanced head and neck squamous cell carcinoma may require highly morbid surgery. We evaluated short-course neoadjuvant anlotinib plus paclitaxel-cisplatin as a signal-generating strategy.
METHODS
Twenty-one patients received two 21-day cycles in a prospective, single-arm Pha...

Background Neoadjuvant chemoradiotherapy (NCRT) is standard for locally advanced esophageal squamous cell carcinoma (ESCC), while neoadjuvant immune-chemotherapy (NICT) is a promising alternative. This study compared pathological response and long-term survival between NICT and NCRT followed by surgery using a target t...

PURPOSE
We assessed the feasibility and efficacy of a response-adapted surgical strategy in patients with locally advanced head and neck squamous cell carcinoma (LA-HNSCC) achieving major tumor regression after neoadjuvant immunochemotherapy (NAICT).
