TL;DR

Serum GDF-15 was associated with an exploratory count of affected clinical domains in adults with IIMs, and no statistically significant correlations were detected with creatine kinase or the Medical Research Council sum score.

Abstract

Objective

Idiopathic inflammatory myopathies (IIMs) are systemic autoimmune disorders with diverse clinical manifestations. Growth differentiation factor-15 (GDF-15) is a stress-responsive cytokine whose clinical associations in IIMs remain incompletely characterized. We examined serum GDF-15 and its relationships with clinical and laboratory features in IIMs.

Methods

This single-center cross-sectional pilot study included 37 patients with IIMs and 25 healthy controls. Between-group differences were assessed by the Mann-Whitney U test and age- and sex-adjusted linear regression of log-transformed GDF-15. Within the IIM group, associations were assessed using Spearman correlations. The principal analysis examined the association between GDF-15 and an exploratory count of involved cutaneous, bulbar, pulmonary, and cardiac domains using partial Spearman correlation adjusted for age, body mass index, and estimated glomerular filtration rate (eGFR).

Results

Serum GDF-15 was higher in patients with IIMs than in controls (median [IQR], 1650 [1110-2739] vs. 544 [344-1006] pg/mL; p < 0.001; adjusted geometric mean ratio, 2.56; 95% CI, 1.93-3.38; p < 0.001). GDF-15 was associated with a greater number of clinical domains involved (ρ = 0.451, p = 0.005); the adjusted partial correlation was 0.408 (95% CI, 0.143-0.662; p = 0.017). No statistically significant correlations were detected with creatine kinase or the Medical Research Council sum score.

Conclusion

Serum GDF-15 was associated with an exploratory count of affected clinical domains in adults with IIMs.

Serum growth differentiation factor-15 levels in Behçet’s syndrome

Behçet’s syndrome (BS) is a heterogeneous systemic vasculitis characterized by chronic inflammation and immune dysregulation. Growth differentiation factor-15 (GDF-15), a member of the transforming growth factor-β superfamily, has been implicated in chronic inflammatory and immune stress responses. This study aimed to...

Association between serum levels of growth differentiation factor-15 and rheumatoid arthritis activity

Growth differentiation factor-15 (GDF-15), also known as macrophage inhibitory cytokine-1, is a member of the transforming growth factor-beta superfamily involved in macrophage regulation. Its exact role in inflammation and tissue repair remains unclear. To evaluate GDF-15 as a biomarker for rheumatoid arthr...

Relapse in adult idiopathic inflammatory myopathies: clinical predictors and the potential role of anti-NXP2 in a single-center retrospective cohort study.

Younger age, anti-NXP2 positivity, and IVIG use independently increased relapse risk, whereas long-term maintenance therapy substantially delayed relapse, highlighting the need for long-term immunosuppression and close monitoring of high-risk subgroups.

FABP3, a Candidate Biomarker of Muscle Strength and Myositis Disease Activity in Patients with Idiopathic Inflammatory Myopathies

Background: Identifying novel, reliable blood-based biomarkers is essential to better assess disease activity in idiopathic inflammatory myopathies (IIMs). We aimed to determine serum fatty acid binding protein 3 (FABP3) levels in IIM patients and evaluated their correlations with disease activity, muscle strength, and...

Serum GDF15 adds prognostic value to forced vital capacity in fibrotic interstitial lung disease.

BACKGROUND Antifibrotic therapy slows fibrotic interstitial lung disease (ILD) progression. However, fibrotic ILD remains a major life-threatening condition, with associated morbidity and mortality increasing annually. Growth differentiation factor 15 (GDF15), a stress-responsive cytokine within the transforming growth...
