TL;DR

Low sorting voltage and recovery regimens were varied and monodisperse spore-suspension preparation protocols and droplet incubation time were optimized for each organism, which revealed that actinomycete-generated droplets were significantly more stable than those of filamentous fungi, with optimal sorting windows dictated by strain-specific growth and morphological traits.

High-throughput droplet screening of filamentous fungi on solid substrates

This work demonstrates a strategy to robustly encapsulate and retain spores of filamentous fungi within water-in-oil droplets by incorporating solid substrates into the droplet microenvironment and overcomes a key technical barrier in applying droplet microfluidics to filamentous fungi.

A Scalable Droplet Microfluidic Platform That Overcomes Large‐Cell Constraints for Long‐Term Plant Cell Culture and Embryogenesis Analysis

ABSTRACT Microspore‐derived embryogenesis (MDE) is essential for doubled haploid (DH) plant breeding, but conventional bulk culture systems are limited by poor scalability and difficulty for automation. Adapting droplet microfluidics for plant cell culture is promising but challenging due to the large size and fragilit...

Application of a microdroplet array platform for high-throughput analysis of growth and natural product biosynthesis of Streptomyces

Streptomycetes are prolific producers of natural products, accounting for the majority of clinically used antibiotics and other bioactive compounds. Their remarkable biosynthetic diversity and ecological versatility are reflected in the large number of biosynthetic gene clusters (BGCs) within their genomes. However...

Throughput and compartmentalisation shape bacterial recovery in droplet- and plate-based cultivation of hydrogen-oxidising communities

Isolation methods are essential for recovering microbial diversity for physiological characterization. Droplet-based cultivation frequently shows greater taxonomic recovery than conventional plating, but the basis of this advantage remains unclear. Here, we used hydrogen-oxidising enrichment communities as a tractabl...

High-Throughput Dual-Optical Droplet Screening for Quantitative Functional Bacterial Selection

Droplet microfluidics enables ultra-high-throughput biological screening, yet most platforms rely on single-parameter optical detection, limiting the information available for selection and analysis. Here, we present a high-throughput droplet screening and sorting platform that integrates simultaneous fluorescence an...
