This paper systematically summarizes the molecular regulatory mechanisms of ferroptosis in HCC, including abnormal alterations in core pathways such as iron metabolism, lipid peroxidation, and glutathione peroxidase 4 (GPX4), as well as its regulatory effects on tumor proliferation, metastasis, and the immune microenvironment.
Abstract
: Hepatocellular carcinoma (HCC) is [1] the most common and most lethal primary liver cancer globally, characterized by complex pathogenesis, difficult early diagnosis, frequent therapeutic resistance, and poor overall prognosis. As a novel iron-dependent form of programmed cell death, ferroptosis has attracted extensive attention in the field of tumor biology in recent years. Meanwhile, this review focuses on the potential value of ferroptosis in reversing therapeutic resistance in HCC. In addition, this paper also summarizes the application prospects of ferroptosis-related gene-based biomarkers in the diagnosis and prognostic evaluation of HCC, as well as the latest research progress of ferroptosis-targeting therapeutic strategies (such as GPX4 inhibitors, system xc⁻ inhibitors, iron n anomedicines, etc.), providing new ideas and potential intervention targets for the precise treatment of HCC.
Hepatocellular carcinoma (HCC), the most prevalent primary liver malignancy, is characterized by a rising global incidence, dismal prognosis, and high mortality rates attributed to its complex etiology and therapeutic resistance. Sorafenib, the first approved systemic therapy for advanced HCC, is limited by the rapid e...

Purpose: Hepatocellular carcinoma (HCC) is the most common primary liver cancer, accounting for nearly 90% of cases and causing over 700,000 deaths annually. Increasing evidence indicates that dysregulated lipid metabolism plays a critical role in hepatocarcinogenesis by promoting tumor initiation, progression, metasta...

Colorectal cancer (CRC) remains a global health concern and a leading cause of cancer mortality, where therapeutic resistance persists as the central barrier to sustained response. Most conventional therapies rely on apoptosis, but CRC frequently resists through p53 alterations, anti-apoptotic BCL-2 upregulation, and d...

Pancreatic cancer is a highly aggressive malignancy with a poor prognosis and pancreatic ductal adenocarcinoma (PDAC) constitutes the overwhelming majority of histological subtypes. KRAS mutation-driven metabolic reprogramming renders PDAC highly dependent on metal ion homoeostasis and antioxidant systems, thereby prov...
