In vivo, knockdown of circARID1A inhibits tumor growth in subcutaneous xenograft models and promotes GC cell proliferation, migration, and invasion by regulating the miR-147a/SNAI2 axis.

Abstract

ABSTRACT Due to the late stage at which most gastric cancer patients are diagnosed, the prognosis is often poor. Current treatments have limited efficacy for advanced cases. This study delves into the involvement of circARID1A and the mechanisms behind its role in gastric cancer progression. GES-1 cell line and GC cell lines HGC-27, AGS, MKN45, and SNU-16 were cultured and subjected to various assays. RNA interference and overexpression techniques were used to modulate circARID1A and miR-147a levels in vitro, while xenograft tumor models in BALB/c nude mice were used to assess tumor growth in vivo. Cellular growth, migration, and invasion were assessed using CCK8, colony formation, and Transwell assays. Dual-luciferase reporter assays, qPCR and Western blot were utilized to investigate the interactions between circARID1A, miR-147a, and SNAI2. CircARID1A promotes GC cell proliferation, migration, and invasion by regulating the miR-147a/SNAI2 axis. In vivo, knockdown of circARID1A inhibits tumor growth in subcutaneous xenograft models.

CircENO1 drives gastric cancer progression via SPI1-mediated transcriptional activation of ENO1.

BACKGROUND Circular RNAs (circRNAs) are critical regulators of tumor biology. However, the role of circENO1 in gastric cancer (GC) remains unclear. This study investigated the biological function and molecular mechanism of circENO1 in GC. METHODS CircENO1 expression was evaluated using a publicly available dataset, c...

MiR-128-3p Inhibits Esophageal Squamous Cell Carcinoma Progression by Targeting TTC9

Background: MicroRNAs (miRNAs) serve as targets for cancer diagnosis and therapy and have the potential to regulate the occurrence and progression of Esophageal squamous cell carcinoma (ESCC). Tetratricopeptide repeat domain 9 (TTC9) gene may involve in the invasion and metastasis of cancer cells. This study aimed to i...

MicroRNA-2110 acts as a tumor suppressor in gastric cancer via direct targeting of IGF1R

MicroRNA-2110 has been implicated in cancer, but its expression and function in gastric cancer (GC) remain understudied. This study aims to explore the role of miR-2110 in GC and its regulation of IGF1R. RT-qPCR was used to measure miR-2110, IGF1R expression in 138 paired GC and adjacent normal tissues. Ex...
