Findings identify HOTAIR as an important molecular regulator of BMSC fate determination and provide mechanistic insights into ceRNA-mediated regulation in SONFH pathogenesis.

Abstract

Background Steroid-induced osteonecrosis of the femoral head (SONFH) is characterized by disrupted osteogenic and adipogenic differentiation of bone marrow mesenchymal stem cells (BMSCs). The long noncoding RNA HOTAIR is aberrantly upregulated in SONFH-BMSCs, yet its functional role in the differentiation imbalance remains incompletely understood. In this study, we investigated whether HOTAIR silencing could restore the differentiation balance in SONFH-BMSCs and clarify the underlying regulatory mechanism. Methods HOTAIR was knocked down in SONFH-BMSCs using small interfering RNA (si-HOTAIR). Osteogenic differentiation was assessed using alkaline phosphatase (ALP) and Alizarin Red S (ARS) staining, while adipogenic differentiation was evaluated using Oil Red O staining. Expression of lineage-specific markers and Wnt pathway components was measured using qRT-PCR and Western blotting. The interactions among HOTAIR, miR-217, and DKK1 were validated through dual-luciferase reporter and rescue experiments. Results HOTAIR silencing significantly enhanced osteogenic differentiation, as evidenced by increased ALP activity, mineralization, and upregulation of RUNX2, BMP2, and Osterix, while concurrently suppressing adipogenic differentiation, with reduced lipid droplet formation and downregulation of PPARγ, C/EBPα, and Adipsin. Mechanistically, HOTAIR acted as a candidate competing endogenous RNA (ceRNA) by sponging miR-217, thereby relieving miR-217-mediated repression of DKK1 and inhibiting Wnt/β-catenin signaling. HOTAIR knockdown restored miR-217 availability, decreased DKK1 expression, and enhanced β-catenin expression. Conclusion HOTAIR inhibition rebalances osteogenic and adipogenic differentiation of SONFH-BMSCs via the miR-217/DKK1/Wnt/β-catenin axis. These findings identify HOTAIR as an important molecular regulator of BMSC fate determination and provide mechanistic insights into ceRNA-mediated regulation in SONFH pathogenesis.

A nucleus-associated miR-661-C/EBPα-PPARγ regulatory axis skews BMSC lineage commitment in SONFH.

Steroid-induced osteonecrosis of the femoral head (SONFH) is characterized by marrow adiposity, trabecular deterioration, and impaired bone repair, but the progenitor-level regulatory mechanisms underlying this process remain incompletely defined. Here, we investigated whether dysregulated bone marrow stromal cell (BMS...

Regulatory role of lncRNA DANCR in osteogenic differentiation of bone marrow mesenchymal stem cells through targeting the miR-19a-3p/PTEN/AKT/mTOR pathway.

Investigation of lncRNA DANCR's role in osteogenic differentiation of bone marrow mesenchymal stem cells and underlying mechanisms found it potentiated the OD capability of BMSCs, as evidenced by increased ALP activity, calcium nodule formation, and elevated expression of OPN, RUNX2, and OCN.

BIRC3 facilitates osteogenic differentiation of bone marrow mesenchymal stem cells via NGFR/β-catenin signalling axis

Aims Osteoporosis primarily results from age-related declines in bone marrow mesenchymal stem cell (BMSC) differentiation into osteoblasts; however, the regulatory mechanisms remain unclear. Methods Integrated analysis of Gene Expression Omnibus (GEO) and RNA sequencing (RNA-seq) data identified a key osteogenic regula...

NAT10-mediated ac4C modification of BMP6 regulates osteogenic differentiation of bone marrow-derived mesenchymal stem cells in the pathogenesis of Adolescent idiopathic scoliosis.

BACKGROUND Adolescent idiopathic scoliosis (AIS) is a three-dimensional spinal deformity. Bone morphogenetic protein 6 (BMP6) drives osteogenesis; this study probes BMP6 mechanism and its potential role in the impaired osteogenic differentiation of AIS-BMSCs. METHODS AIS-BMSCs were isolated and their surface markers...

R-spondins suppress adipogenic differentiation in fibro-adipogenic progenitors via autocrine WNT/β-catenin signaling

Fibro‐adipogenic progenitors (FAPs) are muscle‐resident stromal cells that support skeletal muscle regeneration and homeostasis but can drive pathological adipogenesis. In this study, we investigated the role of R‐spondins (RSPOs), a family of WNT signaling modulators, in FAP adipogenic differentiation. FA...
