Findings indicate that the ORF2-based ic-ELISA is a reliable, cost-effective, and high-throughput tool for seroepidemiological investigation and post-vaccination monitoring of GAstV-1 in geese.
Abstract
Goose astrovirus genotype 1 (GAstV-1) is a major causative agent of visceral gout in goslings, leading to substantial economic losses across Chinese poultry farms. Currently, no commercial vaccine is available, rendering early serological surveillance essential for disease control. In this study, a sensitive and specific indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) was developed using a monoclonal antibody (mAb) targeting the ORF2 capsid protein of GAstV-1. Following optimization of antigen coating concentration, serum dilution, mAb concentration, and incubation time, the assay demonstrated a detection limit at a serum dilution of 1:160 and showed no cross-reactivity with antisera against other avian pathogens, including H9N2 avian influenza virus (H9N2-AIV), avian reovirus (ARV), goose parvovirus (GPV), goose astrovirus genotype 2 (GAstV-2), and Tembusu virus (TMUV). The cutoff percent inhibition (PI) value was established at 25%, based on 40 GAstV-negative sera. Intra- and inter-assay coefficients of variation (CV) ranged from 0.41-0.88% and 3.12-6.95%, respectively, confirming excellent reproducibility. Evaluation of 35 clinical serum samples revealed an 88.6% (31/35) concordance with virus neutralization tests (VNT), with a kappa value of 0.77. These findings indicate that the ORF2-based ic-ELISA is a reliable, cost-effective, and high-throughput tool for seroepidemiological investigation and post-vaccination monitoring of GAstV-1 in geese.
Ganjam virus (GANV) is a tick-borne zoonotic arbovirus that has been reported in India. It is the Asian variant of Nairobi sheep disease virus (NSDV), which causes significant morbidity and mortality in small ruminants. Sero-surveillance for GANV-specific antibodies among animals is relevant for identifying risk ma...
Chlamydia felis is a primary cause of feline conjunctivitis and upper respiratory tract disease. Detection of C. felis largely relies on molecular tests; however, these tests cannot assess prior exposure or herd immunity. In China, no standardized commercial serologic assay for C. felis is available. We developed an in...

Introduction Avian metapneumovirus subtype C (aMPV/C), a re-emerging and widespread epidemic in China, causes hydrosalpinx fluid syndrome (HFS) in laying ducks, resulting in significant economic losses to the poultry industry. In the absence of any licensed vaccines or antiviral drugs for aMPV/C, accurate diagnosis and...

A highly specific blocking ELISA was developed for serological diagnosis of porcine epidemic diarrhea (PED) using monoclonal antibodies (mAbs) against the S1 protein of PEDV. The S1 protein was expressed in HEK293F cells and used to immunize BALB/c mice. Through hybridoma screening, a high-affinity monoclonal antibody...

Rotavirus A (RVA) causes severe diarrhea in donkey foals, but rapid quantitative antigen assays for vaccine development and manufacturing control are lacking. We developed a VP6-based antigen-capture ELISA (acELISA) for donkey-origin RVA using rabbit anti-RVA polyclonal IgG as the capture antibody and horseradish perox...

Toxoplasma gondii is an important zoonotic parasite affecting humans and livestock, and reliable serological tools are needed for disease surveillance in small ruminants. This study developed and evaluated a recombinant T. gondii matrix antigen 1 (TgMAG1) based indirect ELISA (iELISA) for detecting anti T. gondii antib...
