Abstract

Abstract Background The medial prefrontal cortex (mPFC) has a well-established role in extinction learning, triggering plasticity and cellular inhibition via projections to the amygdala. Simple behavioral models such auditory threat (fear) conditioning demonstrate how the infralimbic (IL) mPFC inhibits amygdala output during extinction learning, which is a key component of cognitive behavioral therapies for anxiety disorders. Aims & Objectives A simple modification of threat conditioning, in which a platform allows the rat to avoid the shock, permits the study of active avoidance decisions, where the animal must integrate threat with the pursuit of rewards. We aimed to uncover the circuits involved in active avoidance and its extinction. Method Using a combination of pharmacological interventions, neural recordings, and optogenetic tools, we studied the involvement of the prelimbic mPFC (PL), amygdala, and ventral striatum in rats engaged in platform-mediated avoidance. Results We demonstrate that the PL is crucial for suppressing avoidance behavior during extinction learning. The PL integrates contextual and emotional information from hippocampus and orbitofrontal cortex, and modulates behavioral responses by projecting to the ventral striatum, a region involved in action selection and motivation. Discussion & Conclusions These findings identify a key neural pathway that could be targeted to reduce pathological avoidance in anxiety-related condition such as PTSD and OCD.

221. Investigation of the neural circuitry underlying the extinction of inhibitory avoidance memory in mice

Abstract Background The inhibitory avoidance (IA) task is widely employed as a translational preclinical model for aversive memory and post-traumatic stress disorder (PTSD). IA paradigms recapitulate core PTSD-like phenotypes, including persistence, context-specific avoidance and resistance to extinction. However, the...

Good vibes keep worries at bay: a prefrontal-hypothalamic pathway engages positive affect to mitigate fear

This study shows that key cortico-hypothalamic projections manage extinction retrieval and positive affect to promote fear extinction, and discovered that SIRT1-expressing mPFC-SuM extinction-recruited projections control extinction relapse vulnerability.

694. Amygdala circuit mechanisms modulating emotional memory strength and extinction

Abstract Background Environmental cues present during threatening situations become linked to danger. When these cues are encountered again, they trigger retrieval of a fear memory via specialized neural circuits that produce internal fear states and initiate defensive behaviors. In brain areas such as the basolateral...

618. Modulation of hippocampal fear engrams reveals circuit mechanisms linking maladaptive fear, memory deficits, and depressive-like behaviour

Abstract Background Classic PTSD symptoms have been linked to maladaptive fear memory processing, involving fear generalization and impaired extinction learning. In contrast, cognitive and mood symptoms remain understudied, and their link to fear-related mechanisms is unclear. Aims & Objectives Here, we investigate the...

A ventral hippocampus-prefrontal circuit mediates the pro-extinction effects of adult hippocampal neurogenesis on contextual fear.

BACKGROUND Adult hippocampal neurogenesis (AHN) contributes to hippocampal functions including contextual information processing and stress responses. Inhibiting AHN increases vulnerability to stress and hinders effects of therapeutic interventions for stress-related disorders, especially post-traumatic stress disorder...

Dopamine D2/3 receptors and functional connectivity during fear extinction: A PET-fMRI study

Background The ability to learn what is dangerous in the environment and what no longer exerts a threat is essential for survival. The latter process is studied using Pavlovian fear extinction, the repeated presentation of a feared stimulus that gradually attenuates the fear response. Dysregulation of the extinction pr...
