Age- and sex- related patterns in hematobiochemical and anthropometric parameters of Macaca monkeys.

Abstract

Nonhuman primates are widely used in preclinical biomedical research, with hematobiochemical values serving as key clinical pathology parameters for assessing health status. In this study, we collected and analyzed 2973 blood samples from 566 cynomolgus macaques and 193 rhesus macaques, along with anthropometric measurements such as BMI, body weight, and waist circumference. Corresponding human data were obtained from the NHANES database and incorporated into the study. Regarding white blood cell composition, neutrophil percentages increased while lymphocyte percentages declined with age, with relatively sharp changes observed during both the early stage near sexual maturity and the late stage near aging onset in our macaque and human datasets. The inflammatory indices, calculated based on these compositions, showed similar age-related trends, reflecting age-associated immune modulation in primates. The inflection points approximately correspond to the ages of sexual maturation and aging onset. The statistical analyses predicted a divergence: human aging is primarily defined by metabolic syndrome caused by excess waste accumulation, whereas aging in cynomolgus macaques is fundamentally linked to nutrient deficiency and physical frailty. Despite this contrast, our findings also revealed convergent aging markers across primates. In summary, this study establishes reference indices for hematological and biochemical parameters in macaques, providing valuable insights into age-related changes in primate blood profiles.

Urine and Serum Proteome and Lipidome Analysis of Naturally Aging Feline Species.

Aging in companion animals such as cats closely relates to human aging in environmental exposures and disease manifestation, providing a valuable model for identifying biomarkers of age-associated decline. This study provides a combined proteomic and lipidomic analysis of serum and urine from naturally aging domestic cats aged 3.8-16 years, grouped as adult, old, and senior, to identify age-related molecular changes across biofluids. Label-free quantitative proteomics identified 901 urinary and 238 serum proteins, with 75 urinary proteins significantly altered with age that are linked to kidney disease, hypertension, neurodegeneration, and metabolic disorders. In contrast, only six serum proteins differed significantly between adult and old/senior cats, including decreased Apolipoprotein A-I (APOA1) in seniors, a protein linked with cognitive function in aging. Untargeted lipidomics revealed increases in specific serum triacylglycerols, phosphatidylcholines (PCs), and sphingomyelins, while urinary lipid profiles showed limited age-related changes, with some PCs decreasing, and diacylglycerols increasing with age. These results demonstrate distinct systemic and renal molecular remodeling during feline aging and highlight the utility of integrated omics analyses of biological fluids for identifying molecular alterations relevant to both feline and human aging.

Blood-Based Biomarkers for the Preterminal Stage Using Rhesus Macaques Exposed to Acute Total-Body Radiation

Acute radiation syndrome is characterized by progressive hematologic decline and biochemical imbalances, yet the transition from recoverable injury to the preterminal state remains incompletely characterized using routine laboratory assays. Prior proteomic, metabolomic, and transcriptomic studies have revealed biological signatures in preterminal serum and plasma samples from irradiated nonhuman primates, suggesting that the preterminal state represents a distinct phenotype. In this study, complete blood count and serum biochemistry data collected from male and female rhesus macaques (Macaca mulatta) exposed to total-body doses of 60Co γ-radiation were monitored from seven days prior to irradiation through 60 days post-irradiation. Preterminal samples were collected immediately prior to euthanasia upon determination of moribundity. A series of analyses was performed to characterize differences in injury severity and recovery between survivors and non-survivors. Our findings showed shared early cytopenic decreases in both survivors and non-survivors, consistent with expected radiation-induced hematopoietic injury. Preterminal samples collected between days 14–22 post-irradiation were characterized by erythroid decline and lymphocyte depletion. Most notably, red blood cells, hemoglobin, and hematocrit showed a delayed decline, with significant decreases associated with non-survival. Serum chemistry data was less discriminatory during scheduled sampling, but preterminal comparisons demonstrated significant changes prior to mortality. These results suggest that conventional clinical metrics can help characterize the preterminal phenotype.

Reference intervals for hematology and plasma biochemistry and comparison with selected estimation methods in Northern boobooks (Ninox japonica ) undergoing rehabilitation

The findings underscore the need for caution when interpreting the estimated values in clinical settings and because rehabilitated individuals may appear clinically stable while harboring subclinical pathologies, the results of this study should be considered within the context of rehabilitation.
