TL;DR

A taxonomic appraisal of Microsorum in Niue is presented using chloroplast and nuclear DNA sequence data obtained from 25 samples along with an assessment of morphological characters and the habitats occupied, indicating complex hybridisation involving M. grossum, M. papuanum, and M. scolopendria.

Abstract

We present a taxonomic appraisal of Microsorum in Niue using chloroplast ( rbcL , rps4 + rps 4‐ trn S, trnL + trn L‐ trn F) and nuclear ( gapCp ) DNA sequence data obtained from 25 samples along with an assessment of morphological characters and the habitats occupied. The genetic data collected from the Niue samples were analysed along with sequence data from GenBank based on previous studies of Microsorum in the Asia–Pacific region. Chloroplast data confirmed that M. grossum , M. membranifolium and M. scolopendria are present in Niue as monophyletic clades were recovered for each. The nuclear gapCp data revealed variation comprising both divergent paralogous loci and allelic diversity within loci, with some combinations shared among samples, indicating complex hybridisation involving M. grossum , M. papuanum and M. scolopendria . For some samples, their morphology, habitat and field identification did not correspond with their genetic characterisation, consistent with hybrid origin. Flow cytometry estimates of 2C DNA content (49.16–50.82 pg) did not provide evidence of recent allopolyploidy, indicating that variation observed in gapCp does not reflect differences in chromosome number. Notes on the species habitats and recognition in Niue are presented, including a dichotomous key.

Investigation of the genetic diversity of pearl spot, Etroplus suratensis (Bloch, 1790), through DNA barcoding and phylogenetic analysis from two habitats

Etroplus suratensis (pearl spot) is a commercially important cichlid native to South and Southeast Asia, yet its genetic variation in habitats of Karnataka has not been previously studied. This study integrates morphological and molecular approaches to examine specimens from freshwater (n=65) and brackish water (n=60)...

Species diversity of the Amolops monticola group (Anura, Ranidae) in southern Yunnan, China

Due to morphological conservatism, taxonomic misidentification has frequently occurred in the Amolops monticola species group. In this study, based on previously published data and newly collected samples, phylogenetic and morphological analyses were conducted to investigate the species diversity of the A. montico...

Crenubiotus yalini sp. nov. (Eutardigrada: Adorybiotidae) from China discovered through integrative taxonomy

Based on morphological and morphometric data (obtained with the use of light contrast and scanning electron microscopy) as well as on molecular phylogenetic analyses (based on nucleotide sequence data of 18S rRNA, 28S rRNA, ITS-2 and COI), this study reports a new tardigrade species, Crenubiotus yalini sp. nov., from t...

DNA Barcoding-Based Evaluation of Genetic Diversity in Acanthodactylus (Daudin, 1802) (Lacertidae: Squamata) Species of Türkiye

DNA barcoding of the mitochondrial COI gene is applied to assess genetic diversity and phylogenetic relationships of four species distributed in Türkiye to confirm the validity of all four species and provide the first COI barcode reference library for Turkish Acanthodactylus.

Molecular assessment of confiscated Sceloporus lizards (Squamata: Phrynosomatidae) reveals unresolved taxonomic affinities and genetic reference gaps in Honduras

Molecular tools play a crucial role in biodiversity research and wildlife forensics, yet Central America remains underrepresented in genetic reference databases, particularly for reptiles. In Honduras, species of the genus Sceloporus are poorly studied despite their ecological relevance and increasing demand in the...

Revisiting the Genus Mariosousa (Fabaceae, Mimosoideae) with Complete Taxon Sampling and New Molecular and Morphological Evidence

Mariosousa , a genus segregated from Acacia sensu lato due to morphological and genetic distinctions, is hypothesized to be monophyletic but requires further investigation with expanded taxon sampling and integrated analyses. This study aimed to test the monophyly of Mariosousa , evaluate its infrageneric relation...
