The findings indicate that GmAK1 is a multifunctional regulatory factor and a promising candidate gene for breeding programs aimed at enhancing soybean resistance to phosphorus starvation and nutritional value.
Abstract
Global soybean production is severely limited by phosphorus (P) deficiency, yet the underlying genetic factors linking low-phosphorus adaptation to seed composition traits have not been elucidated. In this study, transcriptomic analysis revealed that Aspartate Kinase 1 (GmAK1) expression is upregulated in leaves under phosphorus-deficient conditions, with the phosphorus-inefficient variety Tiefeng 3 (K5) exhibiting higher mRNA levels than the phosphorus-efficient variety Liaodou 13 (K4). Phenotypic analysis indicated that, under phosphorus-limiting conditions, overexpression (OE) lines exhibited lower total root length, surface area and volume compared to the wild-type control, whilst root phosphorus uptake efficiency was reduced and individual plant yield also decreased significantly. In contrast, the ak1s were able to maintain root volume and yield per plant during phosphorus deficiency, suggesting that GmAK1 overexpression alters key processes involved in root structural reorganisation and impairs the ability to efficiently absorb phosphorus. Furthermore, haplotype analysis of 581 soybean lines identified two major haplotypes, with Hap1 exhibiting higher seed oil content compared to Hap2. This association was functionally supported by the observation that OE seeds contained significantly lower oil content than the wild-type. In summary, our findings indicate that GmAK1 is a multifunctional regulatory factor and a promising candidate gene for breeding programs aimed at enhancing soybean resistance to phosphorus starvation and nutritional value.
BACKGROUND
Nitrogen is the most abundant mineral nutrient required by rice (Oryza sativa) and the primary limiting factor for grain yield. Glutamine synthetase (GS) is a key enzyme involved in nitrogen assimilation in rice, and is closely associated with nitrogen use efficiency (NUE) and source-sink nitrogen dynamics....

Phosphorus (P) deficiency severely limits agricultural productivity in tropical acid soils. Stylosanthes guianensis (stylo), a vital forage legume in the tropics, exhibits high tolerance to low P stress. While numerous phosphate (Pi) starvation response genes have been documented in stylo, its underlying regulatory mec...
It is indicated that overexpression of GmJAZ5 enhances salt tolerance in soybean by strengthening antioxidant defenses, limiting Na+ translocation, and modulating phytohormone homeostasis, highlighting its potential as a candidate gene for breeding salt-tolerant soybean varieties.

Phosphorus (P) is an essential macronutrient required for plant growth and productivity, and its uptake and distribution are mediated by members of the phosphate transporter (PHT) family. Among them, the transporter PHT2.1 has been implicated in chloroplast phosphate homeostasis; however, its role in coordinating plant...
Locule number, a key agronomic trait determining tomato yield and quality, is regulated by genetic determinants, low temperature, and nitrogen supply, and is also affected by phosphorus concentration during the seedling stage, although the underlying mechanism remains unclear. To address this, we treated ‘Mati’ tomato...
Introduction Soybean productivity is highly sensitive to heat and insufficient nitrogen supply, stresses that can occur simultaneously. However, organ-specific lipid responses to this stress combination remain poorly resolved. Methods We exposed hydroponically grown soybean plants to nitrogen deficiency, heat stress (H...
