TL;DR

It is demonstrated that c-Myc protein stability differentiates the response of Ras-mutant MM cells to Ras and MEK inhibition, and suggested that direct targeting of the Ras oncoprotein, rather than its downstream MAPK pathway, may present a more effective strategy.

Dual inhibition of JAK and MEK suppresses T-cell acute lymphoblastic leukemia induced by mutant IL7R and RASGRP1 overexpression.

Gain-of-function mutant IL-7Rα (mutIL7R) occurs in approximately 10% of patients with Tcell acute lymphoblastic leukemia (T-ALL), an aggressive pediatric cancer. Nearly half of patients with T-ALLs also overexpress RAS guanyl nucleotide exchange factor (RASGRP1), an activator of RAS. Since mutant RAS itself has been sh...

MAPK-driven MYC/p21 axis controls senescence bypass and BCL-XL dependence in vincristine-resistant rhabdomyosarcoma

Summary Therapy resistance remains the primary cause of treatment failure in rhabdomyosarcoma (RMS), the most common pediatric soft tissue sarcoma, yet underlying mechanisms are poorly understood. Using vincristine-resistant fusion-negative RMS models, RNA sequencing, genetic perturbations, and a syngeneic zebrafish tu...

Small-molecule RAS/RAF inhibitors target RAS-driven cancers via allosteric RAF disruption

RAS, a frequently mutated cancer-driver gene, has been the focus of intensive research, with numerous inhibitors developed to target RAS and its signalling molecules. With the advent of approved subtype-specific RAS inhibitors targeting KRASG12C mutation, the development of RAS inhibitors that are effective regardless...

Vulnerability of triple negative breast cancer to daraxonrasib pan-RAS(on) inhibition

Inhibiting pan-RAS(on) RAS-GTP with daraxonrasib, alone or in combination with a MEK inhibitor, provides a new strategy for the treatment of TNBC that inhibits RTK activation of the RAS-RAF-MEK-ERK pathway and tumor proliferation.

Targeting Distinct Cell Cycle Nodes Overcomes RAS Inhibitor Resistance.

Activating mutations in KRAS drive pancreatic ductal adenocarcinoma (PDAC) and non-small cell lung cancer (NSCLC). Although mutant-selective KRAS inhibitors and pan-RAS inhibitors provide clinical benefits, the development of resistance limits durable responses. Here, transcriptomic and proteomic analyses revealed that...
