TL;DR

A surface-marker framework based on CD55, CD319, and CX3CR1 is developed that accurately identifies transcriptionally defined human CD8 T cell populations using standard flow cytometry and establishes a biologically grounded framework for CD8 T cell classification and provides a practical platform for mechanistic studies, biomarker discovery, and cellular immunotherapy applications.

Molecular heterogeneity and clonal origin of CCR8+ effector regulatory T cells in human cancer

CD4+CD25+FOXP3+ regulatory T cells (Treg) are highly activated in tumors and promote disease progression. Specific, universal targeting of these effector Treg cells is limited by the lack of a conserved signature across human cancers and information on their origin. Here we combine analysis of single-cell RNA-sequencin...

Phenotypic Alterations of CD8+ and CD8− CD56+ NK Cell Subsets in Moderate and Severe COVID‐19 Disease

The COVID‐19 pandemic has created a global health challenge. Severe cases are associated with immune system dysfunction, which can lead to uncontrolled inflammation. There are no published data on the specific roles of natural killer (NK)‐cell subsets and immune checkpoint (IC) molecules in disease severity. Thirty‐fiv...

Restricted TCR Diversity and Expansion of CD177+CD8+ and TLR4+CD4+T Cell Subsets in Septic Shock‐Associated Immunosuppression: A Single‐Cell Multi‐Omics Study

Sepsis‐induced immunosuppression contributes to poor outcomes in sepsis patients, yet the mechanisms underlying T cell dysfunction remain incompletely understood. We integrated single‐cell RNA sequencing (scRNA‐seq), paired single‐cell T cell receptor sequencing (scTCR‐seq), bulk RNA sequencing (RNA‐seq), and flow cyto...

Elevations of Th17.1 Cells (CXCR3+ CCR6+ T Cells With IL-17A and IFN-γ Production) in Peripheral Blood of Patients With Lung Cancer.

Peripheral T cell profiles reflect antitumour immunity, yet systemic immune shifts often remain confounded by demographic factors. This study aimed to delineate true tumour-driven T cell alterations in lung cancer. We analysed peripheral blood from 121 lung cancer patients and strictly age- and sex-matched healthy cont...

HLA-II Expression Marks Activated and Clonally Expanded CD8+ T Cells and Improves Tumor-Reactivity Prediction Across Human Cancers

Tumor-infiltrating CD8+ T cells are transcriptionally heterogeneous, and robust features linking their clonal dynamics to experimentally defined tumor reactivity remain incompletely characterized. We integrated single-cell RNA sequencing (scRNA-seq) and paired T-cell receptor sequencing (scTCR-seq) data from 31 single-...
