Understanding Chiral Chromatography: With Focus on Mechanism and Factors Influencing Enantioseparation

Aug 2026 · Current Topics in Chemistry · 0 citations

Abstract

Chiral chromatography is a crucial technique for separating enantiomers and plays a vital role in the pharmaceutical industry for drug development and quality control, as many biologically active compounds are chiral. This study examines the underlying mechanisms governing enantioseparation, focusing on key interactions such as hydrogen bonding, Van der Waals forces, and steric effects. Enantiomer separation begins with their binding to the chiral stationary phase, where their unique three-dimensional structures lead to varying affinities and retention. This paper explores key factors influencing enantioseparation, including chiral stationary phases, solvent polarity, temperature, and mobile phase composition. Selection of a chiral stationary phase is vital for method development and influences enantioseparation. This paper sheds light on various available chiral stationary phases, their composition, and the mechanism involved in enantioseparation of various enantiomers in the reported literature. By understanding these mechanisms, researchers can optimize conditions for effective enantioseparation, thereby advancing applications in pharmaceuticals, food safety, and environmental analysis. This review provides an overview of the complex interplay of variables, which guides the researcher in developing efficient chiral separation techniques.
