GENETIC ORGANIZATION OF BLUETONGUE VIRUS AND ITS SIGNIFICANCE FOR VACCINE DEVELOPMENT

Abstract

Bluetongue virus (BTV) is a classic member of the genus Orbivirus and one of the most genetically complex arboviruses infecting ruminants. Its segmented, double-stranded genome, comprising ten RNA segments, provides exceptional capacity for genetic reassortment, high variability, and rapid adaptation to new ecological and epidemiological conditions. Over the past two decades, advances in whole-genome sequencing and comparative genomics have enabled the identification of new BTV lineages, the characterization of numerous reassortants, and the clarification of evolutionary relationships between serotypes. These data have enabled the identification of key protective genes and epitopes associated with the development of effective immunity, as well as the elucidation of molecular determinants of virulence, transmissibility, and adaptation to various Culicoidesspecies. This review summarizes current data on the genetic organization of BTV, its phylogenetic structure, and global evolutionary trends. Particular attention is paid to the genetic mechanisms of pathogenesis, the role of mutations and reassortment in the formation of new phenotypes, as well as methods for mapping antigenic determinants using NGS, bioinformatics, and reverse genetics. Promising areas for the development of next-generation genetically based vaccines are discussed, including multiserotype and DIVA-compatible platforms based on the analysis of protective genes and epitope variability. Вирус блютанга (BTV) является классическим представителем рода Orbivirusи одним из наиболее генетически сложных арбовирусов, поражающих жвачных животных. Его двуцепочечный сегментированный геном, включающий десять РНК-сегментов, обеспечивает исключительную способность к генетической реассортации, высокой вариабельности и быстрой адаптации к новым экологическим и эпидемиологическим условиям. За последние два десятилетия, благодаря развитию полногеномного секвенирования и сравнительной геномики, были выявлены новые генетические линии BTV, описаны многочисленные реассортанты и уточнены эволюционные связи между серотипами. Эти данные позволили идентифицировать ключевые протективные гены и эпитопы, связанные с формированием эффективного иммунитета, а также установить молекулярные детерминанты вирулентности, трансмиссивности и адаптации к различным видам Culicoides. В обзоре обобщены современные данные о генетической организации BTV, его филогенетической структуре и глобальных эволюционных трендах. Особое внимание уделено генетическим механизмам патогенеза, роли мутаций и реассортации в формировании новых фенотипов, а также методам картирования антигенных детерминант с использованием NGS, биоинформатики и обратной генетики. Рассмотрены перспективные направления разработки генетически обоснованных вакцин нового поколения, включая мультисеротипные и DIVA-совместимые платформы, основанные на анализе протективных генов и вариабельности эпитопов.
