Biology

TL;DR

It is demonstrated that the Vibrio lateral flagellar filament is extensively modified by pseudaminic acid and that Maf is required for filament formation, the first structural characterization of a glycosylated Vibrio lateral flagellar filament and a close link between flagellin glycosylation and motility organelle assembly.

FlgJ cell-wall hydrolyzing activity enhances, but is not required, for flagellum assembly in Salmonella enterica

Salmonella can assemble functional flagella, even when the acetylglucosaminidase activity of FlgJ is genetically inactivated, showing that early steps in flagellar assembly are more diverse than previously thought and that differences in cell wall structure between gram-positive and -negative bacteria likely determine...

Flagellar toxicity: Flagellar synthesis is lytic for Bacillus subtilis in the absence of PBP1.

Flagella are large transenvelope nanomachines but how they transit the peptidoglycan in gram-positive bacteria is poorly understood. A recent model suggested that flagellar basal bodies diffuse in the membrane and become captured at locations in the peptidoglycan with a pore diameter that could accommodate the axle-lik...

Structural and Functional Interrogation of Active Streptococcus pneumoniae Sortase A

Sortase A (SrtA) enzymes covalently anchor surface proteins to Gram-positive bacterial cell walls, promoting colonization and virulence. In Streptococcus pneumoniae, previous studies identified both a domain-swapped dimer and an active refolded monomer, but the active enzyme has not been characterized at the structural...

Mycobacteriophage Adephagia gp48 inhibits essential host cell wall biosynthesis enzyme mannose-1-phosphate guanylyltransferase

It is common for bacteriophages to encode proteins that are strongly inhibitory to growth of the bacterial host, and about 10% of mycobacteriophage-encoded proteins have this property. Adephagia is a Cluster K1 mycobacteriophage and prior cloning and expression of 66 Adephagia non-structural genes identified 14 that ar...

Structure of NO16, a marine non-tailed vibriophage with an unusual symmetry-mismatched vertex arrangement

Non-tailed phages remain underexplored in marine environments, as tailed phages have long dominated sequence and culture collections. Yet recent surveys suggest that non-tailed phages may be more abundant and have distinct impacts on microbial mortality and gene transfer. Here, we solve the structure of Vibrio anguilla...

Conserved residues in the CHASE3 domain of BmsA are involved in motility and biofilm formation in Pseudomonas alkylphenolica KL28.

The bmsA gene of Pseudomonas alkylphenolica KL28T encodes a conserved protein involved in motility and multicellular biofilm formation. Sequence analysis revealed that BmsA contains an extracellular CHASE3 sensory domain, suggesting a role in environmental signal perception. To investigate its function, a CHASE3 domain...
